Objective: Gastric ulcer(GU) is a common digestive system disease inthe world, and the rates of incidence and recurrence vary dramatically indifferent countries and regions. It is generally acknowledged that theoccurrence of gastric ulcer is connected with the abnormal gastric mucosabarrier, the dysfunction between nervus and humoral function, the imbalancebetween factors of local damage and defense. Because of the complicatedpathological mechanism and western medicines limited to symptomatictreatment, it is important to explore effective prescription of traditionaChinesemedicine for the treatment of gastric ulcer. To analyze the mechanism ofHuazhuojieduyuyang decoction on gastric ulcer, we recorded biochemicalindicators and pathological changes in gastric ulcer rat of turbid-toxin model,which provide basis for the clinical application and improve the rate of curinggastric ulcer.Method: After fed7days adaptability60male rats were randomlydivided into blank control group(group A), model group(group B), ranitidinegroup(group C), high dose group of traditional Chinese medicine(group D),moderate dose group(group E) and low dose group of Chinese traditionalmedicine(group F) according to the table of random number, and each grouphas10rats. The rats of group A were raised in the normal environment, givennormal eating and drinking until the end of the experiment. Rats of other fivegroups were housed in high fever and wet artificial climate chamber, fed withhigh fat and sugar diet for14days. Experimental groups were givenanhydrous ethanol in the form of1ml:100g, and then continued giving high fatand sugar diet in the hot and humid environment for7days. After the modelfully formed, experimental groups were anew fed on normal diet and givencorresponding drug intervention. Group A and group B were given0.9% sodium chloride solution. Group C were given ranitidine suspension. GroupD、E and F were given the corresponding traditional Chinese medicinesolution. Each rat was given2ml/d fluid for4weeks, and then we observedthe mental state, hair colour and lustre, activities, excreta character, eating,drinking, and weight of the rats in different period. After drug intervention, therats were fasted for24h. Blood samples were collected to measure serum IL-8,NOS and plasma6-Keto-PGF1α. Then the whole stomach was fast paunchedto measure the ulcer index, and a little stomach tissue was paraffin-embedded、sliced and then HE staining. Tissue morphology of gastric ulcer was observedby an optical microscope.Result:1The general condition of the rats: The mental state of the group A wasvery good, and the hair of all the rats was clean, white and bright. The eating、drinking and the excreta of rats was nomal, of course the weight was graduallyincrease. The wight of other groups with high fat and sugar was more rapidlyincreased than group A. After the modeling, rats became apathetic, eatingsignificantly decreased and weight loss gradually. Given drug intervention, therats of each treatment group had good spirit. The fur color, active state, foodintake, and weight of rats were improved too. Group D improved obviouslyand was closest to group A. Group E was similar with group C.2Histopathology of gastric mucosa: Gastric mucosa of group A wasintegrated and smooth, and there was no inflammatory tissue, edema andhyperemia, and it also had not intestinal metaplasia and atypical hyperplasia.Gastric ulcer of group B was being in the active stage, and there was lots ofinflammatory necrotic exudate, congestion and edema obviously in the gastricmucosa. Compared with group C, the damaged degree in group D was lighter,and the ulcer area was repaired significantly. Group E was similar with groupC with moderate injury. There was visible large ulcer, congestion and edemaobviously in group F.3Plasma6-Keto-PGF1α: The levels of plasma6-Keto-PGF1α in group Awas higher than group C, E and F(P<0.05). Group D was higher than group A, but there were no significant differences. Group D, E and F was higher thangroup B(P<0.05). Group D was higher than group C. there were statisticaldifferences (P<0.05). Group E was higher than group C, but there were nosignificant differences.Serum IL-8: The serum concentration of IL-8in group B、C、D、E and Fwas higher than group A(P<0.05), and group B was higner than group C、D、E and F (P<0.05). Group C was higher than group D (P<0.05)and E (P>0.05),but it was lower than group F(P<0.05).Serum NOS: The levels of serum NOS in group A was higher than groupB, C, E and F (P<0.05). Group C, D and E was higher than group B. Group Cwas lower than group D and E, but it was higher than group F (P<0.05). GroupD was higher than group F (P<0.05), there were statistical differences.Conclusion:1Huazhuojieduyuyang decoction could improve rats’ mental state, haircolor, activity and weight, etc. And they had a certain dose-effect relationship.2High dose of Huazhuojieduyuyang decoction could effectively improvethe levels of serum NOS’s and plasma6-Keto-PGF1α’s expression thanwestern medicine, and could rapidly reduce the level of serum IL-8’sexpression. In addition, it could promote tissue hyperplasia and repaired,protect the integrity of the gastric mucosa, and accelerate ulcer healing.
