ObjectiveTo investigate the impacts of BI2536on the activity of Plkl and the cell proliferation of adrenocortical carcinoma SW13cell line.MethodExpressions of Plk1protein of SW13cell at the different concentrations of BI2536and at the different times were assayed by Western-blot. Cell proliferation of SW13cell at the different concentrations of BI2536and at the different times were detected by flow cytometry (FCM).Resultwe find that the Plkl protein of SW13is reduced gradually and the SW13cell number which were arrested at G2/M phase increase (There is significant difference among the groups) when we increase the concentrations of BI2536within40ug/l. we also discover that the Plkl protein of SW13is reduced gradually and the SW13cell number which were arrested at G2/M phase increase (There is significant difference among the groups) when we extend the time of BI2536on SW13cell at the same concentration.Conclusion1.The mechanism of BI2536lead to SW13cell apoptosis may be the inhibition of Plk1protein, resulting in the SW13cell cycle arrest in the G2/M phase, mitosis cannot proceed.2.The studies have shown BI2536can inhibit proliferations of SW13cell in low level concentration in vitro. The further study about the safety and efficacy of BI2536in xenograft models is needed. |
