| Objective:The aim of this study was to compare the differences between Bio-Oss/simvastatin composite and simple Bio-Oss repair material in the repair of rabbit mandibular alveolar bone defect; Observe the effect of oral simvastatin on the bone healing in surgically created defects in rabbits mandibles; Compared the effect of different way to simvastatin application in the alveolar bone defect repair and orthodontic tooth movement; Examine the influence of simvastatin on orthodontic tooth movement after the alveolar bone defect repair and alveolar bone defect.Methods:24New Zealand white rabbits, established the rabbit mandible critical bone defect model.24New Zealand white rabbits were randomly divided into3groups: Group A:the simvastatin composite Bio-Oss repair the defect; Group B:Bio-Oss repair the bone defect, given simvastatin5mg/kg/d after operation; Group C:Bio-Oss repair the defect area, control group. A, B, and C groups are covered by the Bio-Gide collagen membrane.2rabbits of each group were humanely killed4,8,12weeks postoperatively.12weeks after operation, the remaining2rabbits from each group were forced the lower second premolar to move into bone defect.8weeks later,2rabbits of each group were humanely killed. The graft area was scanned by X-ray and CBCT, quantitative analysis was determined by KaVo3D Vision, histological observation, and paired t test and ANOVA were performed for statistical analysis with SPSS19.0software package.Results:1. Established bone defect (6mm×5mm×8mm) after pull out the first premolar in rabbit mandibular. After12weeks no new bone repaired in experimental group.2. After4weeks,8weeks,12weeks observation of X-ray and CBCT imaging. Bone mineral density measured values of A, B group were lower than in group C (p <0.05). Between the A and B group significant difference (p>0.05). Histological observation Bio-Oss bone meal degradation rate of A, B group faster than the group C. The percentage of new bone measurements show A, B group C group, the difference was statistically significant (p<0.05). A, B group between no significant difference (p>0.05).3. Group A and C orthodontic tooth movement distance is greater than in group B (p<0.01), with a significant difference. Group A and group C was no significant difference (p>0.05).Conclusion:1. Rabbit mandibular can established critical bone defects (6mm X5mm X8mm)after pull out the first premolar.2. The local composite simvastatin and oral simvastatin has the effect of promoting absorption of Bio-Oss bone meal, has a role to promote new bone formation in the bone defect repair.3. After simvastatin composite Bio-Oss and simple Bio-Oss repair bone8weeks later, the lower second premolar is able to move into bone defect.4. After orally applied simvastatin significantly inhibited orthodontic tooth movement. |
