Part1Effect of pretreatment of SAHA on early survival rates during lethal hemorrhagic shock on rats first arrival at high altitudeObjective To investigate the therapeutic effect of SAHA pretreatment of lethal hemorrhagic shock on first arrival at high altitude in rats.Methods32male SD rats were randomly divided into three groups:Sham group (Group Sham),pretreated with normal saline group (Group NS), pretreated with SAHA group (Group SAHA). Rats of group Sham were surgically manipulated but not hemorrhaged. Rats of other groups were reproduced as models of lethal hemorrhagic shock in rats first entering high altitude. Rats of group NS and group SAHA were administered subcutaneously with NS (0.25ml) or SAHA (7.5mg/kg dissolved in0.25ml NS),24hours and immediately before lethal hemorrhaged. MAP were monitored constantly. Arterial blood gas were measured at baseline, end of hemorrhage and60min after shock. Rats were observed180min, record death time point of every rats, decide survival rates of60min,120min and180min of each group.Results Compared with group NS, group SAHA got a higher survival rates of60min,120min and180min (P<0.05).The MAP in SAHA group was significantly higher than NS group from15min to60min after hemorrhaged. Pretreated with SAHA increased arterial blood levels of PH. PaO2and BE,but decreased levels of lactic acid (P<0.05).Conclusion Pretreatment with SAHA increases early survival rates of rats first arrival at high altitude suffered lethal hemorrhagic shock, maintains a stable hemodynamic condition. Rats of group Sham were surgically manipulated but not hemorrhaged. Rats of other groups were reproduced as models of lethal hemorrhagic shock in rats first entering high altitude. Rats of group NS and group SAHA were administered subcutaneously with NS (0.25ml) or SAHA (7.5mg/kg dissolved in0.25ml NS),24hours and immediately before lethal hemorrhaged. Rats were observed180min, then sacrificed (rats not survived to180min were sacrificed immediately before death), measure the lung wet-dry weight ratio (W/D), observe the lung histological injury to obtain the lung injury score. The lung SOD activity,the lung concentrations of MDA, the activity of lung MPO, the lung concentrations of interleukin-10(IL-10),the lung concentrations of tumor necorsis factors-a (TNF-a), TNF-a/IL-10ratio and the expression of Ac-H3K9in various experimental groups were detected.Results The lung histological injury of SAHA group was slighter than NS group. Compared with NS group, the lung injury scores,the lung W/D, the lung concentration of MDA, the activity of lung MPO, the lung concentrations of TNF-a and TNF-a/IL-10ratio were significantly decreased (P<0.05).But lung SOD activity, the lung concentrations of IL-10and the expression of Ac-H3K9increased in SAHA group (P<0.05). Conclusion Pretreatment with SAHA alleviates the lung injury of rats. The probability mechanism is pretreatment of SAHA can induce acetylation of histone, inhibit significantly oxidative stress and reduce the inflammatory effect in the lung.
