A Comparative Study Of Bushen Huoxue Recipe And Yiqi Buxue Recipe On Hematopoiesis Regulation Of Myelosuppression In Mice

Objective To investigate the effect of Bushen Huoxue(BSHX) Recipe and YiqiBuxue (YQBX) Recipe on myleosuppression induced by cyclophosphamide(CP)in mice, and compare their different mechanisms on hematopoietic stem/progenitor cells and hematopoietic microenvironment. This study will providebases to determine the best scheme of traditional Chinese medicine (TCM), aswell as improve curative effect of TCM in prevention and treatment ofmyleosuppression after chemotherapy.Methods130SPF grade ICR mice were used and randomly divided into5groups with the same number of males and females: normal control group,model group, BSHX group, YQBX group and combination group. Treatmentgroups were orally administrated decoctions respectively from the first day tothe end. The experimental model of myleosuppression was established byadministered CP intraperitoneally at a concentration of100mg/kg body weightat day8for3consecutive days. Five groups of animals (10mices/group) wererandomly taken for peripheral hemogram on day7(the day before the CTXinjection), and1,3,7days after the successful model set. On3days and7daysafter CP injected respectively, animals (8mices/group) were sacrificed toinvestigate organ indexes, spleen and bone marrow histopathology, numbers ofbone marrow cells, and serum Interleukin6levels; to flowcytometric analyzeCD+34cells and cell cycle phase，to immunohistochemical detecte cyclin D1,and performe the culture of bone marrow stromal cells(BMSC).Results1.Both of BSHX Recipe and YQBX Recipe can promote the hematopoietic recovery from CP injured, and combination Recipe showedsynergistic effect, which significantly elevated WBC, PLT, RBC and HB, andenhance WBC ’s recovery stably.2..Both of BSHX Recipe and YQBX recipe could alleviate the decrease in bonemarrow hematopoietic cells of cyclophosphamide, and BSHX group was better.Besides, BSHX recipe can ameliorate bone marrow sinusoids congestion.Compared with the model group, three treatment groups can significantlyincreased the proportion of CD+34cells, wherein YQBX group was higher on3days after, and combination group was higher at7days after.3. Both of BSHX Recipe and YQBX Recipe can increase the percentage ofG0/G1phase cells. BSHX Recipe is more effective in regulating cycle phasedistribution, not only can avoid excessive proliferation in the early stage, butalso relieve G1phase retardation, thus recover peripheral hemogram stability.4. BSHX Recipe and YQBX can promote the adhesion and proliferation ofBMSC, protect bone marrow hematopoietic microenvironment. Both of themcan decrease the level of IL-6, so as to accelerate the recovery of hematopoieticfunction, especially when combined used.5. BSHX Recipe and YQBX Recipe can increase the spleen index, and protectthe spleen tissue. They can promote the recovery of splenic nodule andlymphocyte numbers, showing protective effect on immune and medullaryhematopoietic functions.Conclusions1.BSHX Recipe and YQBX Recipe can reduce CP caused bonemarrow cells reduction, and it can increase the promoting effect ofhematopoietic recovery when combined used.2. BSHX Recipe and YQBX Recipe can increase the number of hematopoieticstem/progenitor cells, thereby protect hematopoietic potential, andYQBX Recipe was better. At the late stage of the recovery, combination groupwas more effective.3. BSHX Recipe and YQBX Recipe can repair the cell cycle disorders causedby CP, and BSHX Recipe was better. 4. Bushen Huoxue Recipe and Yiqi Buxue recipe have a protective functionagainst myelosuppression, and that its mechanism of action may be related toprotecting bone marrow hematopoietic microenvironment and mediatehematopoietic cytokine levels. Combination group shows better effect.5. Bushen Huoxue Recipe and Yiqi Buxue recipe can protect the spleenhematopoietic and immune function, thus repair hematopoietic function damage.
