Vanillae Fructus,the ripe fruit of Vanilla planifoli Andrews of which hasthe effects of antispasmodic,sedative,anticonvulsant and so on.It containsmany compositions,such as aldehydes, phenols,terpenes,esters,volatileoil,glycoside,and organic acids ect.The Vanillae Fructus raw material contain avariety of aromatic odor chemical compositions,the aldehydes are the mainlyaroma substance.Vanillae Fructus as a natural aromatic plant is widely used inthe food industry,the pharmaceutical industry has broad developmentprospects,but Vanillae Fructus has not yet been received by the ChinesePharmacopoeia,the literature at home and abroad about the quality standardsof Vanillae Fructus have reported less.In order to more effectively develop anduse of the aromatic plants.Therefore,in this paper,the volatile chemicalconstituents,the determination and quality standards of the raw material havebeen studied systematically.Vanillae Fructus is a valuable natural food flavoring,for its full,richaroma and is widely used in medicine,food and other fields.Vanillae Fructusorigin to Madagascar,the topic to the origin of raw material for the study,byusing the method of HS-SPME-GC-MS technology.The factors of the amountof sample,the fibers, extraction temperature,extraction time and desorptiontime were inspected.The experimental program is simple,practical, help to save the resources of plants resources and protection of the ecologicalenvironment.The optimum HS-SPME condition is determined as follows: takeraw material2g in the sampling bottle, insert containing65μm PDMS-DVBextraction fibers of the head of a manual sampler, under90℃heatpreservation30minutes, then under90℃top empty extraction20minutes,remove and immediately insert chromatography injector, take off with3minutes,splitless mode.Using GC-MS method to analyse,28components areseparated and identified. The mainly volatile components is aldehyde,alsocontains aldehydes, phenols, terpenes, acids,esters, ketones,olefin ect.Amongall the components,the content of Vanillin is the highest, about48.28%,followed by Guaiacol(15.54%).These two components occupied63.82%ofthe total contents of the essential constituents.Thin-layer chromatography is introduced as quality identification for thefirst time. The high performance liquid chromatography are adopted for thecontent determination of its Vanillin and Hydroxybenzaldehyde. Thenature,the shape and examination have been studied systemically. The qualitystandard of Vanillae Fructus raw material have been established.In order to improve the quality evaluation system of Vanillae Fructus,thefingerprint atlas research of the Vanillae Fructus has preliminaryconducted.Use the HPLC law, with the methanol-water as the mobile gradientelution,under the275nm wave length,take the Vanillin as the reference,establish fingerprint atlas of10batches of Vanillae Fructus,finally there are4total peaks in the liquid chromatography figures.After the similarity analysisshow that10batches of Vanillae Fructus ’s similarities are high,the correlationis good.These can be used as study the Vanillae Fructus raw material overallcharacteristics of chromatographic fingerprint atlas and provide a reliablescientific basis for the quality control of Vanillae Fructus.This study has not only laid the foundation for the establishment of Vanillae Fructus quality evaluation system,but also provides a scientific basisfor to the development and utilization of the Vanillae Fructus in themedicine,health care products,food and other industries.
