Study On The Relationship Between Expression Of Aurka, SDF-1,as Well As The LGR5and Clinicopathological Characteristics In Gastric Cancer Patients

Objective: To clarify the role of Aurka,LGR5,SDF-1in the formationand development of gastric cancer,we detected the expression of Aurka,SDF-1and LGR5in gastric cancer and paracancerous tissues and analyzedthe relationship between the expression level of Aurka, LGR5,SDF-1andclinicopathological characteristics cancer patients.Methods: gastric cancer and paracancerous tissues of52gastric cancerpatients were collected just after cutting off during the operations.Theexpression values of Aurka, LGR5, SDF-1in these tissues were detected byreal time fluorescence quantitative polymerase chain reaction,(RT-PCR)andimmunohistochemistry.meanwhile the informations consisted of age,gender,carbohydrate antigen19-9(CA19-9)and carcinoma embryonic antigen(CEA)level in peripheral blood and helicobacter pylori,(HP) valuepreoperation,tumor,s location,differentiation,depth of invation,lymph nodemetastasis and stage of the patients were collected.finally,the experimentaldata combined with clinicopathological informations were analysed.Results: The positive rate of Aurka protein in gastric cancer tissues wereobviously higher than paracancerous ones detected by immunohistoche-mistry,which were94.2percent(49/52) and15.4percent(8/52);The positiverate of LGR5protein in gastric cancer tissues were obviously higher thanparacancerous ones detected by immunohistochemistry, which were86.5percent (45/52) and17.3percent(9/52;The positive rate of SDF-1protein in gastric cancer tissues were obviously higher than paracancerous ones detectedby immunohistochemistry,which were94.2percent(49/52) and5.8percent(3/52)Respectively(P<0.05).The expression level of Aurka gene in gastriccancer tissues were significantly higher than paracancerous detected withRT-PCR,between which the average CT value were6.63±1.85and7.20±1.59(P<0.05);however,there was no significant relationship between theexpression of Aurka gene and clinicopathological parameters includingage,gender,and CA19-9, CEA level in peripheral blood preoperation,tumor,slocation,differentiation,depth of invasion,lymph node metastasis,HPinfected,stage of the patients (all P>0.05).The expression level of LGR5genein gastric cancer tissues were significantly higher than paracancerous detectedby RT-PCR,between which the average CT value were7.67±1.80and9.11±2.37(P<0.05);The expression of LGR5gene in HP negative gastriccancer tissues was significantly lower than that of HP positive gastric tissue,both average CT values were8.59±1.44and6.71±1.63,the difference wasstatistically significant(P<0.05);however,there was no significant relationshipbetween the expression of LGR5gene and clinicopathological parametersincluding age, gender,and CA19-9,CEA level in peripheral bloodpreoperation,tumors location,differentiation,depth of invasion,lymph nodemetastasis,stage of the patients (all P>0.05) The expression level of SDF-1gene in gastric cancer tissues were significantly higher than paracancerousdetected by RT-PCR,between which the average CT value were5.24±1.23and6.00±1.35(P<0.05).however,there was no significant relationshipbetween the expression of SDF-1gene and clinicopathological parametersincluding age,gender,and CA19-9,CEA level in peripheral bloodpreoperation,tumors location,differentiation,depth of invation,lymph node metastasis,HP infected,stage of the patients (all P>0.05).Conclusion:(1)Both gene and protein of Aurka,LGR5,SDF-1expressedsignificantly higher in gastric cancer tissues than paracancerous,whichsuggest that Aurka,LGR5,SDF-1might play a significant role in a process ofthe formation and development of gastric cancer as anti-oncogene.(2)Theexpression of LGR5gene in HP-infected gastric cancer tissues wassignificantly higher than that of without HP infection.(3)The expression ofLGR5gene had nothing to do with clinicopathological parameters whichincluded tumor,s location,differentiation,depth of invasion,lymph nodemetastasis,stage,age,gender and CA19-9,CEA level in peripheral bloodpreoperation of gastric cancer patients.(4)The expression of Aurka gene andSDF-1gene had nothing to do with clinicopathological parameters whichincluded HP-infected、tumor,s location,differentiation,depth of invasion,lymph node metastasis,stage,age,gender and CA19-9,CEA level in peripheralblood preoperation of gastric cancer patients.(5)The were not Relationshipbetween the expression levels of Aurka、LGR5and SDF-1gene in gastriccancer tissues with age,sex, and preoperative CA19-9,CEA detectionvalues.(6)There have no correlation betweenAurka、LGR5and SDF-1ingastric cancer.
