Purpose To investigate the antiinflammatory effcet of triptolide onstromal keratitis of rabbit model.Methods The0.1%triptolide eye drops was dispensed,BSS served ascontrol. The stromal keratitis model was established by stromal injection of0.02%LPS thermosensitive gel on right eyes. After24hours, topicaladministration was performed,4times daily for14days. The observation timepionts were day1ã€day3ã€day7and day14after toical administration, and atevery time point, ten Newzealand male rabbits were randomly divided into twogroups, five for each group.These eyes were observed and scored at every timepoint by ophthalmological microscope. The HE staining and immuno-fluorescence staining (NF-Κb and MIP-1) were also performed to analyze theinflammation response at every time point. All datas were analyzed byStudent’s t-test and expressed as mean±standard deviation (SD). Statisticalsignificance was accepted as P=0.05.Results The day1results after topical administration of the two groupsshowed no differences by statistical analysis. But at the time pionts of day3ã€day7and day14, corneas in triptolide group appeared on a lower scoredinflammatory and immune response, in biomicroscope, HE staining andimmunofluorescence analysis, comparing with the control group, P<0.05.Conclusion1%Triptolide eye drops showed off an apparente antiin-flammatory effcet on stromal keratitis of rabbit model. Traditional chinese medicine triptolide might have a prospect on clinical utilization in future, as areplaceable or adjuvant drug of glucocorticoids. |
