The main active ingredients of panax quinquefolium saponin are ginsenosides. Manyresearches show that the ginsenosides are metabolized in vivo into some partly deglycosylatedsaponins and aglycons, which have significant pharmacological activities. However, thesesaponins and aglycons are very rare in Panax plants. Therefore, modifying the structure ofginsenosides is very meaningful.In this thesis, the panax quinquefolium saponin were degraded under the condition of highpressure for the first time.9compounds were isolated and purified from alkaline-degradation.And their structures were elucidated by the physico-chemical properties and spectraltechniques as follows: dammar-20(21),24-diene-3β,12β-diol(Ⅰ),20(S)-Panaxadio(Ⅱ),(20S,24R)-dammar-3-one-20,24-epoxy-6α,12β,25-triol(Ⅲ),20(S)-Protopanaxadiol(Ⅳ),24(R)-Ocotillol(Ⅴ), Pseudo-sapogenin DQ(Ⅵ),24(S)-Ocotillol(Ⅶ),20(S)-Protopanaxatriol(Ⅷ) and20(S)-dammar-25(26)-ene-3β,6α,12β,20-terol(Ⅸ). Amongthem, compound Ⅲ named Pseudo-ginsengenin R1is a new compound which is first obtainedand has not been reported.The results of this experiment showed that deglycosylated aglycons were mainly abtainedfrom the alkaline-degradation products of high temperature and high pressure ofpanax quinquefolium saponin. When the pressure and the ratio of the material were the same,both20(S)-PPD and24(R)-Ocotillol reduced along with the extension of the reaction time.However,20(S)-PPD decreased faster than24(R)-Ocotillol. This results indicated that thestructure of Ocotillol was more stable.The tonifying Yang activity of the alkaline-degradation products of high tempreture andhigh pressure of panax quinquefolium saponin(QSRP) on the aged male rats was also studiedduring the experiment at the same time. The results showed that the alkaline-degradation couldevidently improve sexual function of senile male rats, including promoting the secretion oftestosterone, improving the NO content in the blood, increasing the contents of dopamine and5-hydroxytryptamine in the hypothalamus compared with blank groups. The results provide a preliminary theoretical basis for aphrodisiac activity of QSRP.
