| Herba Patriniae is a perennial herbal. It is widely separated in China with variouskinds and it has crucial pharmacological and health care function. The present study wasundertaken to access the best extraction process, as well as to analysis the concentrationof flavone in herba Patriniae under different growing conditions and in differentposition. Anti-oxidant and anti-bacterial of flavone from herba patriniae were alsostudied. This paper has provided scientific proof for maximum utilization of herbaPatriniae, including its deep manufacture process and its application on food,medication and others industries.The main research results were as follows:(1) The conditions of solvent extraction were optimized by single factor andorthogonal experiment: the concentration of ethanol60%, the solid-liquid ratio1:20(g/mL), extraction temperature60℃, extraction time2h, the extraction ratio of totalflavone in this condition was1.82%and the highest extraction ratio of flavone extractedfrom tender leaves of herba patriniae was2.13%(2) The invitro antioxidant activity of extract of herba patriniae was studied bymeasuring the abilities of scavenging free radical, nitrite and anti-oil peroxidation. Theresults showed that flavone had strong activities of scavenging hydroxyl free radical,superoxide anion free radical and nitrite,as well as could inhibit oil peroxide. It had apositive correlation relationship between antioxidant activity and the concentration ofextract. It was better than Vc.(3) D-galactose mouse aging model was build. In the mouse behavior experiment,different concentration of flavone from herba patriniae was added in feed. Mouseweight, food intake, organ weight was observed. At the same time, activity of MDA,SOD, GSH-PX and CAT in both organ tissues and blood serum of mouse was measured.The results showed that flavone from herba patriniae:①could improve mousemovement coordination ability, endurance and new environment adaption ability.②hadno remarkable effect on mouse weight, food intake, and organ weight.③couldsignificantly reduce blood serum and MDA content. Activities of SOD, GSH-Px andCAT in both organ tissues and blood serum were improved.(4) Micratiter plates and enzyme-labeling instrument were used. The inhibitioneffect flavone of herba patriniae on biofilm was evaluated by measuring OD630,bacterial cell adhesion test, aggregometry, and motility experiment. The results showed that flavone had inhibition effect on the formation of gstaphylococcus aureus biofilm.The higher concentration of flavone had the stronger effect. When adding the8mg/mLflavone at48h, the inhibition effect was the most significant. |
