Objectiveto investigate the influence of Rosiglitazone on expression of ATP bindingcassette transporter A1,G1and cholesterol metabolism in THP-1macrophages.MethodsThe experiment into3groups:blank controller group;ox-LDL(100mg/L)group;RG (100mg/L(ox-LDL)+10μmol/L (RG)) group. THP-1cells werecultured.Macrophages were induced from THP-1cell by PMA.Two groups aftermodels of foam cells were built by incubating macrophages with ox-LDL for48h.The last group foam cells were intervened with RG for48h.The levels ofABCA1,ABCG1mRNA and protein expression of each group were observed byRT-PCR and Western Blot;Cellular lipid accumulation was determined by OilRed“Oâ€staining;Each group intracellular free cholesterol and cholesterol esterswere analysed qualitative and quantitative by High performance liquidchromatography.ResultsCompared with ox-LDL group,ABCA1,ABCG1mRNA and proteinexpression of RG group THP-1macrophages were markedly increased(P<0.01).The intracellular lipid accumulation and the contents of total cholesterol,freecholesterol and cholesterol esters of RG group were also significantlydecreased(P<0.01). ConclusionsRosiglitazone can increase the expression of mRNA and proteinABCA1,ABCG1of THP-1macrophages and restrain foam cells formingthrough promoting intracellular free cholesterol outflow and reducing cholesterolesters levels. |
