| ObjactiveTo study the effect of Aidi,A chinese medicine, on growth and apoptosis oflaryngeal cancer cell line Hep-2.MethodsHep-2cells were treated with Aidi at0.4,0.8,1.0,2.0ug/ml concentrationsas experimental groups,0ug/ml as control group.After24ã€48and72hours,MTTassay was evaluated in the cellular proliferation inhibition rate, fluorescencemicroscope was testing in the apoptosis,the cell cycle distribution and thechanges of apoptosis ratio were analyzed by flow cytometry,and real-timefluorescent quantitative PCR was used to detected the mRNA expression ofCyclooxygenase-2in the Hep-2cells.Resultsâ‘ Aidi at different concentration inhibited the growth of Hep-2cells in asignificant time-and dose-dependent manner compared with that incontrols(P<0.05). The growth inhibition rate at0.4ã€0.8ã€1.0and2.0μg/mL for72h were23.08%ã€37.42%ã€43.38%ã€54.48%.â‘¡The typical apoptosischaracter-istics was found in the experimental groups from Fluorescence,and normal cellswas significantly reduced. The control group didn′t observe apoptosis changeand the chromatin in cells evenly distributed.â‘¢FCM showed that Aidi decreased the apoptotic rate of Hep-2cells at the G0/G1phase, increased the apoptoticrate of Hep-2cells at the G2/M phases and induced the apoptosis of Hep-2cells.The apoptotic rates at0.4ã€0.8ã€1.0and2.0μg/mL for72h wereincreased28.62±1.23%increased54.90±2.40%, respectively, which were allsignificantly higher than those of the control group(P<0.01).â‘£The expressionlevel of epoxidase was higher in Hep-2cells than in control72h after treatmentwith Aid. the mRNA expression of Cyclooxygenase-2in the Hep-2cells weredecrease by real-time fluorescent quantitative PCR (P<0.05).ConclusionsAidi inhibits the growth of human Hep-2cells by inducing apoptosis of them.The mechanism is related of the mRNA expression of Cyclooxygenase-2through it is cut... |
