ObjectiveTo investigate the influence and significance of the expression of the coretranscriptional factor for the regulation of upstream signal pathway embryonicstem cell(ESC):Nanog which all-trans-retinoic-acid(ATRA) has influence onglioma cell lines SHG-44,U87MG and U251.MethodsEach cell line was divided into the experimental group(B) which was treatedwith the concentration10umol/L ATRA for5days,and the cells of controlgroup(A)was cultured normally without the treatment of ATRA.Then,bothimmunocytochemistry(ICC) and Reverse Transcription Plus Polymerase ChainReaction(RT-PCR) methods are respectively adopted to detect the expressionof Nanog from the protein level and the mRNA level among the three kinds ofcell lines.ResultsThe result of immunocytochemistry(ICC) shows that positive rates ofNanog protein in glioma cell lines SHG-44,U87MG and U251of the controlgroups are respectively(65.43±1.66)%,(64.81±4.10)%and (63.54±2.06)%,andthe difference was not statistically significant(F=0.155,P=0.858)after dealing bythe analysis software SPSS17.0. And the result of Reverse Transcription PlusPolymerase Chain Reaction (RT-PCR) reveals that the core transcriptionalfacter Nanog mRNA of the three glioma cell lines in the relative content were 0.6368±0.0399,0.6421±0.0637and0.6516±0.0444,and the difference in thesecells was not statistically significant by analysis(F=0.427, P=0.662).However,5days after application of ATRA-induced Nanog protein in the three cell lines,thepositive rates of Nanog protein of experimental groups are respectively(36.48±3.72)%,(35.50±4.04)%and(35.23±3.09)%,compared with the controlgrous,and the differences between the control group cells and the experimentalgroup ones are statistically significant(tSHG-44=13.7681, tU87=15.490,tU251=20.963,PSHG-44=0.000,PU87=0.000,PU251=0.000),and Nanog mRNA in these groups wererelatively0.4583±0.0791,0.2551±0.0793and0.3331±0.0540,compared with thecontrol groups the difference was significant (tSHG-44=7.176,tU87=25.556,tU251=14.673,PSHG-44=0.002,PU87=0.000,PU251=0.000).ConclusionsThe core transcriptional fator of embryonic stem cell(ESC) for the regulationof signal pathway—Nanog,of glioma cell lines which are SHG-44, U87MG andU251is highly expressed, and its expression in these cells can be reduced byATRA-inducd,the degree of malignancy of these cells can be lessened.
