Objective:To study the expression and clinicopathology significance of Nucleolin, Ki-67and CCR7in normal oral mucosa(NOM), leukoplakia(LK) and oral squamous cell carcinoma(OSCC) by tissue array analysis. And find the relationship between their expression and the occurrence of LK and OSCC, offer theoretics and new notion for cancerous potential of LK and early diagnosis and treatment of OSCC.Methods:The expression of Nucleolin, Ki-67and CCR7protein was examined by the immunohistochemical technique(PV) in10NOM,34LPand47OSCC samples, and its relation to multiple clinicopathological factors of OSCC, such as lymph node metastasis, histologic grade and muscle invasin was also analyzed. The experimental data were statistically analyzed by SPSS17.0software.Results:(1) The label of Nucleolin, Ki-67were located in nucleolus and CCR7were located in cytosols or membranes. But Nucleolin also labeled in cytosols and/or membranes of some other tumours cells examined by optical microscope. (2) There were significant difference of Nucleolin, Ki-67and CCR7between NOM, LK and OSCC(P<0.05).(3) The expression of Nucleolin and Ki-67were higher in OSCC with poorer grade of histologic differentiation, and dffferent significantly in histologic grade(well vs moderately/poorly)(P<0.05). The expression of Nucleolin were observably different between NOM and OSCC, NOM and LK, and muscle invasin. And the significant difference expression of Ki-67were between NOM and OSCC, LK and OSCC (P<0.05). There were also significant difference expression of CCR7between NOM and OSCC, LK and OSCC, and regional lymph node metastasis (P<0.05). They are similar between those with sexes, ages, smoking or drinking.(4) The expression of Nucleolin and Ki-67was correlated in positive rate of histologic differentiation grade of NOM, LK and OSCC by Spearman analysis.(P<0.05). But the expression of Nucleolin and CCR7, Ki-67and CCR7was not correlated.Conclusions:Expression of Nucleolin protein may play an important role in progression of OSCC, which was associated with subcellular location. And it was more sensitive in OSCC muscle invasion. Nucleolin has the protential of being a new molecular target, as Ki-67, of cell proliferation. Expression of CCR7may incarnate lymph node metastasis sensitively. The detections of expression of Nucleolin, Ki-67and CCR7were most important significance to cancerous potential of LK and early diagnosis and treatment of OSCC.
