Objective: Dental caries, dental trauma and chronic tooth damage can lead todental pulp lesions. If they don’t receive timely treatment,they can cause pulpitis,periodontitis and pulp-periodontal lesions. So control pulp injury seems veryimportant to repair the damage, the most common is the promotion of reparativedentin formation. This study in order to obtain a better pulp capping material andmaintain pulp vitality after the pulp is damaged. Gene transfer is the latest researchprogress, erythropoietin EPO (erythropoietin, EPO) gene in gel sponge place on ratmolar at the exposed pulp to observe the change of the local odontoblast-like cells,pulp itself reaction, inflammation and reparative formation; we can assess its ability torepair the tooth pulp and clinical feasibility.To explore safer, more effective and adaptto a wider range of keeping vital pulp treatment.Erythropoietin (EPO) is a cytokine and is produced by the liver and adult kidney,and it can promote erythropoiesis. In addition, erythropoietin receptors is expressedon the surface of red blood cells, which is an active glycoprotein secreted by thekidneys, it can stimulate erythropoiesis bone marrow, mother cell proliferation anddifferentiation. Molecular weight of human erythropoietin is about35000, consists166amino.It was found in the19th century and produce recombinant humanerythropoietin, in the1980s it has been widely used in clinical. With variety ofapplication in hematopoietic hormone erythropoietin is wide range of clinicalapplications, but at this stage,its main use of its hematopoietic function can promoteproliferation and differentiation of erythroid prog-enitor cells, the research focus onmyocardial infarction,renal failure anemia and so on.In recent years, because of the discovery of the EPO receptor, a wide range ofresearch on non-hematopoietic function,such as the research hot of EPO can promotebone formation and be directly through the role of bone marrow stromal cells, as well as also can indirectly interactions with the bone by signal pathway ofhematopoietic stem cell. Moreover, it can by BMP, VEGF to promote bone formationand tissue repair. Studies have shown that, BMP can promote the formation ofreparative dentin, and the combination of EPO may be involved in the inflammatoryresponse of the pulp. So we boldly hypothesized that EPO has a potential role in therepair and regeneration of dental pulp, and we hope that it is can be proved by thisstudy.Methods: Establish the model of the rat pulp reparative dentin, adenoviruscarrying the erythropoietin (EPO) gene in gel sponge, and then lay up the rat teeth atthe exposed pulp, and placed in saline gel sponge for control. Normal feeding animals,after3days,7days,14days and28days the animals were killed to produce paraffinthe Organization wax block, HE staining and immunohistochemistry analysis andhistological observation.Results: After surgery of3days, the difference was not significant between theexperimental group and the control group. After surgery of7days,14days and28days, compared with the control group, the experimental group with the EPO the pulptissue was significantly active at exposed pulp. After7days, you can see theinflammatory response lighter than the control group, occasional there is RDformed;14days after the operation, odontoblast-like cells active; after14days and28days, the local organizations of exposed pulp has significantly ability to improvereparative formation.Conclusion: Transfer to the exposed pulp of adenovirus carrying of EPO genecan promote reparative formation and make actively reaction of odontoblast-like cells.It also can reduce necrotic pulp tissue formation. This study obtain expected results.Itcan lay the theoretical foundation-based drugs and methods used in clinical. Moreover,EPO used the pulp tissue can promote pulp regeneration for the first time, which hasobvious innovation.
