| The invasion and distant metastasis of the tumor is a complicatedpathological process, involving the formation of tumor blood vessels andvascular basement membrane and extracellular matrix degradation,etc.RECK gene, is a newly discovered tumor suppressor, which expressedin most normal tissues,but low expressed in the cancer cells. One of themain factors of the invasion and distant metastasis of the tumor is matrixmetalloproteinases, which induced tumor cells to produce proteasedegradation of extracellular mtrix. RECK can inhibit the expression ofMMPs, thus inhibiting the invasion and distant metastasis of the tumor,when the cells have malignant transformation, RECK expression cut ormissing. Up to now, RECK regulates mainly three matrixmetalloproteinases: MMP-2and MMP-9and MTIMMP (MMP-14).Recently, one of the research hot spot is MMPs and their inhibitors.Experimental investigation shows that the expression of RECK inesophageal squamous carcinoma, external auditory canal squamous cellcarcinomas, skin squamous carcinoma, colon cancer, hepatocellularcarcinoma, which is related to the occurrence, development andinfiltration of the tumor, and the expression of RECK often and negatively correlated with the expression of MMPs. However, theexpression of RECK has only a few literature reports in oral squamouscell carcinoma. The purpose of this project is to investigate the expressionof RECK and MMP-9protein and their correlations with the occurrence,development,invasion and metastasis of oral squamous cell carcinoma.Methods SP immunohistochemistry method was used to detect theexpression of RECK and MMP-9protein in59cases of oral squamouscell carcinoma and10cases of oral mucosa. Statistical analysis wasperformed using chi-square test.Results The protein expression of RECK and MMP-9were closelycorrelated with the tumor grade,infiltration and lymph node metastasis inoral squamous cell carcinoma(all P <0.05). The expression rate ofRECK protein was increased in tumor and the oral mucosa,which was44.06%and90%,respectively,and there was significant differencebetween every two groups(P <0.05);however,the expression rate ofMMP-9protein was decreased in tumor and the oral mucosa,which was74.6%,10%,respectively,and there was significant difference betweenevery two group(sP <0.05). The expression rate of MMP–9protein andthe expression rate of RECK, has nothing to do with the age and thegender. The expression rate of RECK protein and the expression rate ofMMP-9are negatively related.Conclusion RECK and MMP-9may play important roles in the infiltration and metastasis of mucosal epithelium in oral squamous cellcarcinoma. United detection of RECK and MMP-9can be used in theearly diagnosis and prognosis judgement of oral squamous cell carcinoma.The high expression rate of RECK protein indicates good clinicaloutcomes, because the high expression of RECK inhibits the expressionof the MMP-2and MMP–9,in order to inhibit angiogenesis and theinvasion and metastasis of the tumor, and improve the prognosis ofpatients. In tumor cells, the expression rate of RECK is low,it suggestswe can through the research of drugs, which can raise the expression ofRECK,inhibiting tumor metastasis. |
