| China has rich plant resources with great varieties. Systematically investigation ontraditional Chinese medicine and folklore medicinal plants is important for the discovery ofnew and active compounds from mother nature, and also that can offer scientific basis forcomprehensive development and utilization for these plants.In this thesis, two folklore medicinal plants named Vernonia nantcianensis (Pamp.)Hand.-Mazz. and Astragalus membranaceus (Fisch) Bge. were chemically investigated,and11compounds were obtained and structures of these compounds were elucidated on1HNMR and13C NMR. Crepiside E (1),3β-[4-(4-hydroxyphenyl)acetyl-β-D-glucopyranosyl-oxy]-8α-hydroxy-4(15),10(14),11(13)-guaiatrien-12,6-olide(2), Crepiside H (3), Youngiaj-aponicoside A (4), β-Sitosterol (5)ã€Taraxasterol (6),9,10-Dimethoxy-pterocarpane-3-O-β-D-glucoside (7), Calycosin-7-O-β-D-glucoside (8), Astragaloside â… (9), Astragaloside â…¡(10), Astragaloside â…£(11). Compound1-6were obtained from Vernonia nantcian ensis(Pamp.) Hand.-Mazz. for the first time.To establish a HPLC-DAD method to determine the content of Crepiside H in Vernonia nantcianensis extract and Astragaloside â… in Astragalus extract to establish qualityspecification.Based on single factor experiment, the best ultrasonic extraction conditions ofsreceiving rate of Astragaloside â… were optimized by Box-Behnken Design in surfaceoptimization method.The interreaction of ctDNA with11compounds obtained from Vernonianantcianensis extract and Astragalus extract has been researched by UV-Visspectrophotometry, the maximum absorption wavelengh of Crepiside E, Crepiside H andCalycosin-7-O-β-D-glucoside had red shift. Calculating the constants of ctDNA withCrepiside E, Crepiside H and Calycosin-7-O-β-D-glucoside, find the constants of CrepisideH were biggest. These phenomenas explain that a bittle interaction of ctDNA withCrepiside E, Crepiside H and Calycosin-7-O-β-D-glucoside has been existed, and moreexperiments need to prove way of the interaction of these active compounds with DNA. |
