Objectives To observe the behavior changes in different periods of pentylenetet-razol-induced kinding rats, and detect the hippocampus5-HT levels after kinding successfully.To explore the effects of different doses of citalopram pre-treatment on pentylenetetrazol-induced kinding rats in latent period and seizures degree and the hippocampa5-HT levels after kinding succufully, to provide a strong experimental evidence and theoretical basis for exploitation new type anti-epileptic drugs.Methods60SD rats, male or female,were divided to6groups in random,blank control group, PTZ model group, PTZ+CTP1group(CTP1mg/kg), PTZ+CTP2group (CTP5mg/kg), PTZ+CTP3group(CTP1Omg/kg), PTZ+CTP4group(CTP20mg/kg)(N=10in each group).Separate intervention by different doses of citalopram(lmg/kg、5mg/kg、1Omg/kg、20mg/kg) intragastric administration once day on9AM before PTZ40mg/kg intraperitoneal injection0.5hour, PTZ model group with an equal amount normal saline, the blank control group all with an equal amount normal saline. Observed of behavior for20minutes and recorded latent period and seizures degree everyday at the1d,5d,10d,of PTZ kingding. Once a day until fully lit. Hippocampus5-HT levels were detected by immunohistochemical technique.Results(1) Blank control group showed no seizure activity, PTZ model group and PTZ+CTPgroups showed seizure activity.(2) Behavioral observationsCompared with PTZ model group.There were a lighter seizures degree and longer latent period in the PTZ+CTP1group, PTZ+CTP2group, PTZ+CTP3group in1,5,10day. The effect of anti-epileptic was depanded on dose-dependent. Among four groups (P<0.05); At the1day, compared with PTZ model group, there was a heavier seizures degree in the PTZ+CTP4group (P<0.05), but the latent had no statistically significant (P>0.05). At the5d、10d, there were a heavier seizures degree and shorter latent period in the PTZ+CTP4group (P<0.05).(3) Hippocampus5-HT levelsCompared with PTZ model group, there were highter hippocampus5-HT levels in the PTZ+CTP1group, PTZ+CTP2group, PTZ+CTP3group,and5-HT content was proportional to amount of CTP dose. Among four groups (P<0.05); Compared with PTZ model group, there was lower hippocampus5-HT levels in the PTZ+CTP4group (P<0.05). Blank control group had highter hippocampal5-HT levels than PTZ model group (P<0.05).Conclusions(1)5-HT is a modulator of epileptogenesis; And its effect on seizure activity dep-end on hippocampus of5-HT levels. Within a certain range, it may play an inhibitory role,and the effect of anti-epileptic was depanded on5-HT content. But when exceeds a certain range, it may show a excitatory effect.(2) Citalopram could regulate seizure activity may by affecting5-HT level. lmg/kg,5mg/kg,10mg/kg citalopram played an inhibitory role. its antiepileptic effect was depanded on CTP dose;20mg/kg citalopram can promote seizure activity.
