| Objective:To observe the content CGRP and SP of plasma, the expression5-HT and c-fos and c-Jun gene in the brainstem of migraine model of rats, and the effect of sinomenine on its content and expression, to discuss on the mechanism of sinomenine on migraine, and to broaden the clinical application of sinomenine provide theoretical basis.Methods:60wistar rats (half female and half male) were randomly divided into6groups:Normal control group, model group, sumatriptan group, sinomenine low-dose intervention group, sinomenine medium-dose intervention group, sinomenine high-dose intervention group. The model of migraine rats were reproduced in accordance with Cristina Tassorelli method, after modeling and drugs intervention, the content CGRP and SP of plasma were measured by radioimmunoassay and the positive cells of5-HT and c-fos and c-jun gene in brainstem were measured by immunohistochemical SP and SABC method, and at the same time the rat’s behavioral changes were observed.Results:①The rats’behavioral observations showed that the intervention group of nitroglycerin-induced migraine behavioral symptoms gradually disappeared with time passing by;②Among the groups there was no significant difference (P>0.05) in the content CGRP and SP of plasma;③Different from the model group and the normal control group, in the sumatriptan group and sinomenine intervention groups of various doses the5-HT-positive cells in brainstem significantly increased, the difference was statistically significant (P<0.01);④In contrast to the normal control group, the c-fos and c-jun expression in the brain stem cells within the model group increased significantly, the difference was statistically significant (P<0.01); compared with the model group, the c-fos and c-jun expression in brainstem cells significantly decreased in the sinomenine intervention groups of various doses and the sumatriptan group, the difference was statistically significant (P<0.01);⑤Compared with the sumatriptan group, the difference in the numbers of5-HT-positive cells in brainstem was not statistically significant (P>0.05) in the sinomenine medium-dose intervention group;⑥There was no significant difference (P>0.05) in terms of the expressions of c-fos and c-jun in brainstem cells between the sumatriptan group and the sinomenine high-dose group.Conclusion:①Sinomenine has the effect of treatment on migraine caused by GTN, in the central exists the site of action for sinomenine analgesia;②Sinomenine achieves the effect of treatment on migraine,the mechanism maybe by raising the activity of5-HT system of brain-stem;the other hand,Sinomenine can inhibit the c-fos and c-jun gene expression in brain stem cells during attacking migraine, and diminish the long response of intracranial pain. |
