Objective:Functional roles in antibacterial peptides were analyzed in Black soldier fly (Hermetia illucens L.) larvae, Using a series of different induced ways. Accordingly, our results could provide the additional types of antibacterial peptides and the parameters for experimental extraction of crude extracting for the antibacterial, antiviral and antitumor drug researches.Methods:Products were harvested from Black soldier fly larvae, Using staphylococcus bacteria liquid injection induction of E. coli and further grinding extraction of crude extracting antimicrobial peptides. In this study, acupuncture soaking induction was applied to obtained antibacterial peptides in the fifth stage of animals involving bacterium fluid and staphylococcus aureus bacteria liquid, associated with diverse frequencies and different time in Black soldier fly larvae. Moreover, grinding extractions of antimicrobial peptides were performed to select for activity analysis of the crude products. And then, biological measurements of crude extractions were carried out after those products were testified by the reversed phase high performance liquid chromatography (RP-HPLC).Results:Just as shown in our studies, the highest quality of antibacterial peptides would be obtained from5years old animal in Black soldier fly larvae. The variable effects were observed in those experimental animals suffered from the different induction method such as E. coli bacterium fluid acupuncture soaking for60s. The highest quality of products were harvested from24h growth after inductions, and then, the following effects were from the high frequency induction with ultrasonic (100W) for20min. Obviously, The worst results, no significant comparison of the control, were detected by the other methods as described above. The satisfied biological activity was measured from cultivations of Black soldier fly administrated to the suitable temperature (for example27,37and50℃) for10min. The products should be discarded from4times freeze-thaw treatments, compared with the satisfied antibacterial peptides of6times freeze-thaw administrations. The decreased activity appeared in these products from the range of pH5.0-9.0, even if the mild antibacterial activity from pH<5.0and pH>9.0. The independent peak of separated antibacterial peptides was validated with reversed phase high performance liquid chromatography (RP-HPLC).Conclusion:1. The highest quality induction of antimicrobial peptides could be obtained from5years old Black soldier fly larvae.2. The differential antimicrobial effects were detected by diverse induction methods. The highest effects were obtained from acupuncture soaking for60s and24h cultivations.3. The satisfied biological activity was measured from cultivations of Black soldier fly administrated to the suitable temperature (for example27,37and50℃for10min. The products of freeze-thaw treatments from no more than4should be harvested.4. Reversed phase high performance liquid chromatography (RP-HPLC) was validated to get the independent peak of separated antibacterial peptides.
