| Ginko biloba polyprenol(GBP) is one kind of lipid component of ginko biloba leaves,whcih contains17~22isopentene group. The chemical structure of GBP is similar to thedolichol in human oranges. The content of GBP in ginko biloba leaves is as high as1%~2%,that is100times higher than the content of dolichol in human organs. Therefore, it is the bestway to seclect GBP for large scale preparation of dolichol. The synthesis of a series ofdervatives of GBP through the derivatization and the further research of the activity providesa foundation for the exploitation and application of GBP, which has important practicalsignificance and broad prospect.The extraction and purification of GBP mainly contains the following steps. Steamdistillation was used to get rid of volatile oil; petroleum ether was used to get the extractwhich was then saponified with5%NaOH-EtOH to prepare the unsaponifiable matter; theheavier component and the light component were obtained after the molecular distillation;urea embedding method was used to purify the heavier component; dilica gel columnchromatography was used for purification. The purity of GBP was determined by the HPLCquantitative analysis, the result showed that in the unsaponifiable matter the purity was22.31%, in the heavier component the purity was37.91%, the purity was72.34%after ureaembedding purification and the purity of the final product was as high as90.22%.The reaction condition was optimized In the research of hydrogenation synthesis ofdolichol with GBP. The result showed that the optimum reaction was2:1(V/Vmethanol:dichloromethane); mass ratio was100:1(m/m GBP: catalyst); the reaction timewas24h; reaction pressure was10MPa, the coversion rate was46.58%. The structure of thehydrogenation puoduct was charactered by HPLC, IR,1H-NMR and13C-NMR.In the research of synthesis of the derivatives of GBP, the reaction conditions wasoptimized and the purification process was epplored. The yield rate of polyprenyl sodiumphosphate was55.18%, the yield rate of polyprenyl acetate was93.50%and the yield rate ofpolprenyl ammonium salt was67.98%. The structure of the products were charactered by IR,1H-NMR and13C-NMR.The diameter of inhibition zone and MIC of GBP and its derviates were determined, theantibacterial spectrum was studied and also investigated the scavenging effect of thegydrogenation product to O2.-and·OH. The result showed that the GBP and its derivativesdisplayed a good antibacterial effect against Sa. and Ec. but the antibacterial effect againstAspergillus and Beer yeast was poor and the antibacterial effect of derviates was superior tothe raw material. Also found that the hydrogenation product had a good antioxidant activity. |
