The expression of level of PPARγ and NF-κB/P65in placenta among the two groups were determined by immunohistochemistry. There were no significant difference in the age, gestational weeks, body mass index, gravid and parity in the two groups. They were diagnosed according to the criteria (Obstetetrics and Gynecology, the Seventh Edition), without complications of internal medicine and other complications of pregnancy. All placentae were collected prior to labour onset, at caesarean section.Results:(1) The expression of PPARγ protein was mainly observed in the nucleus of sycytiotrophoblasts, a few in cytotrophoblasts and the stromal cells. The expression level of PPARγ in patients with preeclampsia was significantly higher than the control group (p<0.01).The expression level in patients with severe preeclampsia was significantly higher than in patients with mild preeclampsia (p<0.05).The expression level of PPAR γ protein in placenta of preeclampsia women with intrauterine growth restriction (IUGR) was significantly higher than those without IUGR(P<0.01). (2) NF-κB/P65was mainly located in placental sycytiotrophoblasts, in vascular endothelial cells and a few in the stromal cells. The expression level of NF-κB/P65in patients with preeclampsia was significantly higher than the control group (p<0.01).The expression level of NF-κB/P65in patients with severe preeclampsia was significantly higher than in patients with mild preeclampsia (p<0.05).(3) Correlation analysis showed that the expression of PPARγ and NF-κB/P65in placenta of patients with preeclampsia had a positive correlation.Conclusions:(1) The expression level of PPARγ protein in placenta of the preeclampsia group was higher than the control group and correlated with the severity of the disease and the growth of fetus.(2) The activity of NF-κB/P65in placenta of the preeclampsia group was higher than the control group and correlated with the severity of the disease.(3) The expression of PPARγ and NF-κB/P65in placenta of the preeclampsia group had a positive correlation and affected each other, which may participate in the pathogenesis of preeclampsia. Further study the role of PPARγ, PPARγ ligands and NF-κB in pregnancy under physiologic and pathologic mechanisms may be useful in early prediction, diagnosis and therapy for preeclampsia. | | Keywords/Search Tags: | preeclampsia, placenta, immunohistochemistry, PPARγ, NF-κB | PDF Full Text Request | Related items |
