| Skin is an important estrogen target. Estrogens play an important role in skin physiologyand pathology process. Estrogens do not only accelerate cutaneous wound healingã€improvethe inflammatory skin disorders, but also offer some degree of protection against skinphotoaging. The effects of estrogen are predominantly mediated through the estrogenreceptors. At the cellular level estrogen improve the proliferation and migration ofkeratinocyte, fibroblast and mastocyte in order to repair skin disorder, avoid from skin cancerand delay skin aging. The differentiation about ER at distribution and expression in tissue canbe attributed to the polymorphism of gene. The estrogen receptor isoforms regulate multiplesignaling pathways. That plays a decisive role at therapy.ERα and ERβ are two distinct intracellular estrogen receptors that belong to the nuclearreceptor superfamily. They express in all layer of skin. ERα36is a new type of membraneestrogen receptors. The Recent study indicated that ERα36-mediated lower-level E2pathwaysare required for preserving bone mass in postmenopausal women who becomeosteoporotic.The previous experiment results showed ERα36express in MCF-7and liver cellsand it can mediate the PI3K/AKT and MEK/ERK pathways to promote cell growth andcanceration. But how does it express and distribute in skin cell that is unknow. We usedwestern blot to confirm ERα36was expressed in skin cell from rats and human. Furtherinvestigating ERα36co-localized with Cav-1on membrane of skin cell byImmunofluorescence.Results:1) ERα36is expressed in the skin of rats and the relationship between amount ofERα36and Caveolin-1is negative correlation, the amount of difference not only in gender butalso in tissue;2) ERα36and ERα66were expressed in fibroblast and the amount of ERα36ishigher than that of ERα66;3) ERα36is expressed in keratinocytes and tissues of skin. Itpartly co-localized on the cell membrane with Caveolin-1. Conclusion: ERα36is expressed in skin cells and the amount of difference in tissue assimilar as other ER isoforms. ERα36localize on cell membrane. It may participate in theestrogen signal pathway mediated by Caveolin-l and play an important role in the process ofestrogen treatment on skin. |
