Objective:To evaluate the effect of Ginkgo leaf capsules in mean defects index in visual field of glaucoma patients with controlled intraocular pressure by placebo-controlled; to analyze the relationship between mean defect index in visual field and the visual electrophysiology P-VEP, and explore the protective mechanism visual function.Method:A randomized, double-blind, parallel placebo-controlled, multi-center clinical trial was conducted. Patient with glaucoma was the main object of this study. They were collected at the Ophthalmology department of the Affiliated Hospital of Chengdu University of TCM, et al.5hospitals nationwide in all. Patients with the corrected visual>0.3who were diagnosed primary open-angle glaucoma and acute angle-closure glaucoma after surgery with intraocular pressure controlled by medicines or surgery (after more than three months, intraocular pressure≤21mmHg) and visual field defect, were enrolled and randomly divided into test group and control group. The test group was treated with Ginkgo leaf capsule, the control group was treated with placebo capsule. All the course of treatment was24weeks. By observing the visual field and visual electrophysiology before and after treatment by Ginkgo leaf capsules, evaluate the effects of mean defects index in visual field on treating patients with controlled intraocular pressure glaucoma. Datas were analyzed with SAS9.1.3and SPSS13.0statistical software.Result:1. General information:285cases were enrolled in all.142cases were in Ginkgo leaf capsules group and143cases were in the placebo group. Compared the baseline information like visual acuity, gender, age. disease duration, degree of visual field defect and visual field before treatment, the difference was not statistically significant (P>0.05).During the trial, patients had good compliance, didn’t violate the combining medication and treatment of the trail.2. Efficacy of visual field:The MD values after treatment reduced forl.04±2.78in Ginkgo leaf capsules group, it reduced for0.10±3.00in placebo-controlled group. The difference was not statistically significant within control group before and after treatment, and it was statistically significant in test group. There was a statistically significant difference between the two groups.3. Efficacy of pattern visual evoked potential:Compared with before treatment, pattern visual evoked potential, specifically the amplitude of P-VEP0.5cpdN145, lcpdN75and2cpdN145significantly improved after treatment in Ginkgo leaf capsules group, the difference was statistically significant. There was no significant difference before and after treatment.Conclusions:1. Ginkgo leaf capsule could improve the visual field in glaucoma patients with controlled intraocular pressure.2. Ginkgo leaf capsule could promote recovery of the pattern visual evoked potential.3. There was a certain relationship between the improvement of visual field and the recovery of P-VEP. The mechanism may be depressing calcium flowing, and thus inhibit cells apoptosis.
