Objective To study the neuroprotective effect of xingnaojing injection on aquaporin-4(AQP4) expressions in rat brain tissue its neuroprotective after intracerebral hemorrhage (ICH).Methods The ICH rat model was induced by injecting blood of itself. Forty adult healthy male Sprague-Dawley (SD) rats were randomly divided into normal saline control group (NSC, n=10), n=10), ICH group(n=10), hirudin treatment group (HIR, n=10), Xingnaojing treatment group (XNJ, n=10). Hematoxylin eosin and toluidine blue staining was performed to examine perihematomal neurocyte morphological changes. The expression of AQP4in perihematomal tissue was detected by immunohistochemistry and Western bloting respectively after72h.Results There was no pathological changes and weak expression of AQP4in NSC group. The expression of AQP4in perihematomal tissue were increased in ICH, while comparing with ICH group, the tissue morphological changes were improved obviously, and the expression of AQP4were also decreased significantly in XNJ group and HIR group (P<0.05).Conclusions The increased expressions of AQP4might induce some brain injury together following ICH, and xingnaojing injection could alleviate neural injuries after ICH by inhibiting the expressions of AQP4.
