Objective: To construct the Affect of Downregulation of expression of p53gene tosaRNA activate p21gene in human kidney cancer cell line ACHN and human bladdercancer cell line5637.Methods: Synthetic the sequence-specific corresponding saRNA fragments of thepromoter with the P21gene, sequence-specific corresponding siRNA fragments ofmRNA with the P53gene,Co-transfected into human kidney cancer cell line ACHNand human bladder cancer cell line5637in vitro, respectively with Lipofecta-minereagent, Realtime-PCR and Western-blot were performed to examine the differenceexpression level of p53and p21in each group.Results:①p21gene expression in human kidney cancer cell line ACHN and humanbladder cancer cell line5637significantly increased.②p53gene expression inhuman kidney cancer cell line ACHN and human bladder cancer cell line5637significantly declined.③when p53gene down regulated in human kidney cancer cellline ACHN and human bladder cancer cell line5637,p21gene expressionsignificantly increased.Conclusion: synthetic saRNA dsP21–322can activate both the p21gene in humankidney cancer cell line ACHN and human bladder cancer cell line5637, when the p53gene is down regulated, regardless of the p53gene is wild-type or mutant, did notaffect the dsP21-322to activate the p21gene.
