Objective: To study about the osteogenesis effect of different concentration of Rapamycinand the synergism with osteogenesic inducers and the Electromagnetic FieldMethods: We isolate the bone marrow mesenchymal stem cells from4-week-oldSprague–Dawley rats(male,60~80g) and culture them. The bone marrow mesenchymalstem cells (from passage three) are transplant into the plates.1) We divide the stem cells intogroups about:Control group, osteogenesis inducers group,1nM Rapamycin group,5nMRapamycin group,10nM Rapamycin group,20nM Rapamycin group and relativeconcentration group add with osteogenesis inducers. After7days,14days and21days ofstimulation, we extract the RNA from relative groups and explore the expression of theRUNX2, BSP, OPN. Then we make plotting and analyse the data;2)divide the cells intofour groups: control group,5nM Rapamycin group, Electromagnetic field group, theRapamycin+EMF group, we extract the proteins and the RNA and use the alizarin red tostain the calcium disposition about7d,14d,21d stimulation respectively. Then we use WBand RT qPCR and alizarin to detect the result.Results: Rapamycin has a promoting osteogenesis effect which is appreciably lower thanthe osteogenesis inducers in the early period but much higher than the control group. The5nM Rapamycin has a best effect than other groups in different concentration;5nMRapamycin has a much better effect than the osteogenesis inducers in the extended periodwhile other groups are apparently lower than the Osteogenesis induces; all groups whichcontains Rapamycin have better osteogenesis effect than the osteogenesis inducers group inthe late period and Rapamycin has a very profoundly promoting synergism effect withosteogenesis inducers during all periods, and the osteogenesis effect is greatly enhanced with stimulating time,5nM Rapamycin has a much more strong effect than other groups inall period;1mT,15Hz,4h/d EMF does not show a distinct osteogenesis effect in the earlyperiod, on the contrary, the EMF does show a strong effect in the middle and late period,the synergism group of Rayamycin and EMF has a much lower effect than the EMF andRapamycin group.Conclusions:1)Rapamycin has a profounding promoting effect of osteogenesis and theeffect is strengthened with stimulation time,2)5nM Rapamycin group is much moreeffective than other Rapamycin groups,3) Rapamycin has a very profoundly promotingsynergism effect with osteogenesis inducers during all periods,4)1mT,15Hz,4h/d EMFdoes not have an obvious osteogenesis effect in the early stimulating period, but is veryeffective in the middle and late period,5)EMF has an antergic effect with the rapamycin onosteogenesis effect.
