Objective:To study the effects of ecdysterone on inflammatory cytokine and endothelial function of experimental type2diabetic rats and its possible mechanismMethods:fifty male wistar rats were randomly divided into4groups: ordinary feed group ten rats(Group D), always use ordinary food to feed, the rest of the40rats were established type2diabetic rat model through high-fat diet plus low-dose STZ injection, then randomly selected30rats from them and Randomly divided into three groups:ecdysterone treatment group10rats(group B), pioglitazone treatment group10rats(group C) and physiological saline treatment group10rats (A组) each group were given ecdysterone5mg/kg, pioglitazone20mg/kg.d and Physiological saline respective all by irrigation stomach once a day for6weeks.Then determined each rat’s fasting blood glucose and blood fat level, determined the serum concentration of TNF-a and MPC-1with ELISA method, determined the expression level of endothelial type of nitric oxide synthase (eNOS) and endothelin-1(ET-1) in aortic endothelial cells with immunohistochemical method. Result:The fasting blood sugar (FBG) of groupB and C was decreased obviously compared with group A (P<0.01),there was no no obvious difference between group B and group C (P>0.05). The plasma TG concentration of group B and C was significantly lower than the group A (P<0.01), the plasma TG concentration of group B(P<0.01) and C(P<0.05) was significantly lower than the group A.The plasma FFA concentration of B andC was significantly lower than the group A (P<0.01), The plasma LDL-C concentrations of B(P<0.01)and C (P<0.05)was below group A.2.The TNF-α serum concentration of group A、B and C was significantly higher than the group D (P<0.01), group B and C was significantly lower than the group A (P<0.01), differences between group B and C was not statistically significant (P>0.05).3. The MPC-1serum concentration of group A、B and C was significantly higher than the group D(P<0.05), group B and C was significantly lower than the group A (P<0.01).4. Endothelin-1immunohistochemical results:There were obviously tan expression in the aortic endothelial cells cytoplasm of groupA (P<0.01), B (P<0.05)and C (P<0.01) compare with D,there was a significant increase in ET expression in endothelial cells of group A, The endothelin-1expression of group B (P<0.01)andC (P<0.05) abated compare with group A,there was no statistical difference between group B and C (P>0.05).5. eNOS immunohistochemical results: Continuous line tan dyeing can be seen in endometrial area especially endothelial cellscytoplasm of groupD、B and C, which means eNOS positive expression.The aortic lining area tan dyeing of groupA weakened dramatically, blank no express area appear in the local place, the difference between group B、C and groupA is statistical significance (P <0.05). There is no no obvious difference between the groupB and C (P>0.05).Conclusion:ecdysterone obviously reduce fasting blood glucose、blood fat and blood plasma FFA level of experimental type2diabetic rats, it also reduce serum concentration of TNF-α and MPC-1, similar with the effect of pioglitazone. It shows that ecdysterone have the effect of endothelial protection and resistance to atherosclerosis.
