Objective:To discuss the effects of immediate asthma reaction in mice exposured to different concentrations of fluoranthene, naphthaleneMethods:120male Balb/c mice were randomly divided into twelve groups (every group=10). The mice in Group C, which was a asthma model group, were given intraperitoneal injections to Ovalbumin(OVA)20ug+A1(OH)32mg, total2ml, at the1、8、15day and given3%OVA+saline inhalation for30minutes on conescutive seven days since22th day. group A, were given intraperitoneal injections and inhaled to normal saline insteading of ovalbumin, step with the C group. Group B was Alcohol control group, the mice of group B were intraperitoneal injected and inhaled with95%ethanol insteading of ovalbumin, step with the C group. Group D was low concentration (50ug/m3) of fluoranthene group. Group E was middle concentration100ug/m3) of fluoranthene group.Group F was high concentration (200ug/m3) of fluoranthene group. Group G was low concentration(50mg/m3) of naphthalene group. Group H was middle concentration(250mg/m3) of naphthalene group. Group I was high concentration(500mg/m3) of naphthalene group. Group J was low concentration(fluoranthene:50ug/m3+naphthalene:50mg/m3) of fluoranthene and naphthalene group. Group K was middle concentration (fluoranthene:100ug/m3+naphthalene:250mg/m3) of fluoranthene and naphthalene group. Group L was high concentration(fluoranthene:200ug/m3+naphthalene:500mg/m3) of fluoranthene and naphthalene group.The mice of group D、E、F、G were exposed to the different concentrations of fluoranthene in2weeks,4h/d. The mice of group A、B、C were exposed to fresh air at the same time. After group D to L exposured to toxicant, then stimulated ovalbumin asthma model. A week later, all groups were sensitized with3%ovalbumin for30minutes.then test the change of airway resistance were observed for15min and counted eosinophils in bronchial lavage fluid and detected the content of VEGF, MMP-9and TIMP-1in lung tissue by using Immunohistochemistry and Finally observe the pathological changes of lung tissue.Results:①A、B group had no Bronchial hyper-responsiveness,so had no statistical significance(P>0.05). The airway resistances in C to L group were higher than in group A、B, they had statistical significance compared with group A and group B(P<0.01). The airway resistances in D to L group were higher than in group C, and E、F、H、I、K、L had statistical significance compared with group C (P<0.05).②In bronchial lavage fluid of A、B group we did not found or see a small number of eosinophils (EOS). In C to L group, EOS accounted for a percentage of total cells were significantly higher than A group and B group, the differences were statistically significant (P<0.01). Compared with the C group, EOS of D、E group accounted for a percentage of total cells were increased, but no statistical difference (P>0.05).③A and B group’s lung tissue vascular of VEGF, MMP-9, and TIMP-1positive expressions were similar, so had no statistical significance. The A group and B group of mice vascular endothelial cells could see weak expression of VEGF, MMP-9, TIMP-1; D to L group mouse’s VEGF, MMP-9, TIMP-1in the lung tissue cell slurry could see different degree of positive expression, and see nuclei shading, so compared with A group and B group, these group were statistically significant (P<0.01). Mix of high concentration group’s (L group) VEGF, MMP-9were more obvious than asthma model group (group C), so it had statistical significance (P<0.05); high concentration group of TIMP-1(group L) is more obvious than in the asthma model group(group C), so its difference was statistically significant (P<0.05); D-L groups of positive expression raised varying degree than asthma model group (C Group), but mixed medium, low concentration groups, and positive expression of high, medium and low concentrations of fluorescent and naphthalene groups had no statistical significance (P>0.05). The ratio of high concentration Mixed group MMP-9and TIMP-1is obviously increased, and MMP-9was positively correlated with VEGF.④The lung pathological section showed that the endothelial cells in A、B group were integrated, and had no denaturation or necrosis, stroma did not see of inflammation cell infiltrate, periviascular did not see edema.Group C can see lots of inflammatory cells were infiltrated around the airway. D, G, J group can see lots of inflammatory cells were infiltrated around tube wall, peripheral vascular mild edema. E, H, K group of epithelial cell shedding, and lots of inflammatory cells were infiltrated, and peripheral vascular edema is obvious. F, I, L group of epithelial cells was falling, and large amounts of inflammatory cells were infiltrated, peripheral edema. Lesion severity was correlated with concentration.Conclusion:1. Naphthalene and Fluoranthene exposure can aggravate asthma airway hyperresp-onsiveness m mice.2. High concentration(naphthalene+fluoranthene) caused VEGF, MMP-9’s expressi-ons raised, and make MMP-9/TIMP-1’s balance disordered, resulting in airway inflammation、airway hyperresponsiveness 3. Inhalation of Naphthalene and Fluoranthene can aggravate asthma speed response in mice.
