Objective:Acute renal obstruction is very common in Urology. It will result in irreversible pathophysiology of renal fibrosis and finally develop into end-stage renal failure without treatment in time. In recent years, study on therapy of renal disease using mesenchymal stem cells (MSCs) has made great progress. There is not a criterion for MSCs transplantation, however, different routes of delivery can affect homing of MSCs, and homing is the precondition of MSCs therapy. Therefore, I used an animal model of reversible unilateral ureteral obstruction (R-UUO) to simulate clinical acute renal obstruction. MSCs labeled with DiR were transplanted into the R-UUO animals through three routes: renal artery, tail vein and subcapsular. Homing of MSCs was evaluated by in vivo imaging system (IVIS). The results may be useful for later research and clinical practice.Innovations:1. There has not been any research on the evaluation of homing efficiency of the three routes;2. Tracking the homing of MSCs using IVIS in UUO animal model is an innovative method.Methods:1. Thirty SPF femal SD rats (4W) were divided into three groups:renal artery, tail vein and subcapsular. Every group contained five rats at each time point;2. Thawed and subcultured the MSCs, and labeled the MSCs with DiR;3. Built up the animal model of R-UUO;4. MSCs labeled with DiR were transplanted into the R-UUO animals through three routes:renal artery, tail vein and subcapsular;5. The whole body and organs in vitro were imaged by IVIS at the1st hour and the48th hour after transplantation.Results:1. The character of MSCs was not affected by thawing, subculturing and labeling. The fluorescence intensity increased with the concentration of DiR-BM-MSCs;2. I successfully set up the rat R-UUO model, and improved the stability and equilibrium of the model;3.I built up and perfected the technique of the three delivery routes;4. The homing efficiency of DiR-BM-MSCs in obstructive kidney was higher than control kidney in the group of tail vein (P<0.05);5. The homing efficiency of the three delivery routes was significantly different at the early stage (P<0.05).Conclusions:1. MSCs have the character of homing to obstructive kidney;2. The homing efficiency of MSCs transplanted through the three routes is:subcapsular, renal artery, tail vein, from high to low;3. Although the transplantation efficiency of the three delivery routes was the same as the results of MSCs homing efficiency, it need to choose a reasonable route according to actual condition in practical application. |
