Objective:To explore the impact of IL-15on on proliferation and differentiation of HaCaT cells, and then uncover the mechanism. Method:(1) HaCaT cells were cultured and treated with various concentration of IL-15for different time. MTT method was used to analyze the proliferation of HaCaT cells. The makers (K1, INV and K5) of HaCaT cells differentiation induced by CaCl2were detected by Real-time PCR and Western-Blot was used to study the protein expression of differentiation markers. Furthernore, we analyzed signaling pathways induced by IL-15in HaCaT cells by Western-Blot, and then used the specific inhibitors to selectively inhibit ERK and AKT phosphorylation to reveal the relationship between the affect of IL-15on the proliferation and differentiation, and these relative signaling pathways. Results:(1) In HaCaT cells, IL-15promotes the proliferation of HaCaT cells at a dose-dependent and time-dependent ways. However, IL-15inhibited the makers (K1, INV and K5) expression of HaCaT cells differentiation induced by CaCl2at mRNA and protein levels. Mechanistic studies show that IL-15increased the phosphorylation of ERK1/2and AKT, and the specific inhibitors of the phosphorylation of ERK1/2(PD98059) or AKT (LY2940002) could antagonize the effect of IL-15on the proliferation and differentiation of HaCaT cells. Conclusion:(1) The IL-15patially depend on the MAPKs-ERK1/2and PI3K-AKT signaling pathways to regulate the proliferation and differentiation in HaCaT cells. |
