Study on the stability of the chronic emotional stress model induced by empty bottle stimulation:According to the chronic emotional stress model induced by empty bottle stimulation which established by Wenjuan Lin. On the basis of the model which given empty bottle stimulation for14days to carry out empty bottle stimulation with wistar male rats for35days that in order to induce the emotional changes and replicate the model of chronic emotional stress. Observing the change trend of the weight, behavior (attacking, exploring and grooming)and open-field score of the rats to examine the stability of the model.2. Replicate the rats sleep deprivation model induced by chronic emotional stress:Applying the method of the empty bottle stimulation for14days to set up the chronic emotional stress model, which combined with the technique of the cortical EEG of the rats, then observed and recorded the changes of sleeping states of the rats. The animal model of chronic emotional stress----sleep disorders could be established finally.3. The studies of the interventional effects of Semen Ziziphi Spinosae on the sleep-deprivated rats induced by the chronic emotional stress:Wistar male rats were given the empty bottle stimulation for14days to set up the chronic emotional stress mode. From the8th day of the empty bottle stimulation, wistar male rats that were chronic emotional stress mode given with Semen ziziphi spinosae decoction (17.5g/kg crudedrug) for7days by intragastric administration, which combined with the technique of the cortical EEG of the rats to observe the interventional effects of Semen Ziziphi Spinosae on the sleep-deprivated rats induced by the chronic emotional stress. Results:1. Study on the stability of the chronic emotional stress model induced by empty bottle stimulation:The rats were given empty bottle stimulation for35days to induce the chronic emotional stress. The experimental results show that the change of rats behavior (attacking, exploring and grooming) in control, physiological and emotional stress groups are constant. Compared with the rats in control and physiological stress groups, the rats in emotional stress group expressed significant attacking behavior, and the rats in control and physiological stress groups expressed exploring and grooming behaviors. Compared with control group, the square crossing, rears, grooming in open-field test and body weight for the rats in emotional stress group had significant differences, while physiological stress group had no significant differences.2. Replicate the rats sleep deprivation model induced by chronic emotional stress:Applying the method of the empty bottle stimulation for14days to lead to the chronic emotional stress and observe the changes of sleeping states of the rats. The experimental results show that compared with control group, the slow-wave sleep (SWS) for the rats in emotional stress group obviously reduced while the light sleep extended comparatively, then the total sleep time (TST) shortened finally. But the rapid eye movement sleep (REMS) had no significant changes.3. The studies of the interventional effects of Semen Ziziphi Spinosae on the sleep-deprivated rats induced by the chronic emotional stress:Chronic emotional stress mode was given with Semen ziziphi spinosae decoction for7days by intragastric administration. The experimental results show that the both slow-wave sleep (SWS) for the rats in drugs given group and the total sleep time (TST) extended. The rapid eye movement sleep (REMS) had no significant changes. Compared with the rats in emotional stress group, the rats in drugs given group that the body weight, attacking behavior, square crossing and rears in open-field test had significant differences. Conclusion:1.Study on the stability of the chronic emotional stress model induced by empty bottle stimulation:The rats model of chronic emotional stress behaviors caused by empty bottle stimulation for35days were constant relatively.2. Replicate the rats sleep deprivation model induced by chronic emotional stress:The chronic emotional stress caused by empty bottle stimulation for14days had significant effect on sleeping states of the rats. The chronic emotional stress could lead to the sleep deprivation in rats.3. The studies of the interventional effects of Semen Ziziphi Spinosae on the sleep-deprivated rats induced by the chronic emotional stress:Semen ziziphi spinosae decoction could improve the sleep deprivation status of the rats by lengthening the slow-wave sleep to extend the total sleep time. Besides, Semen ziziphi spinosae decoction could relieve the attacking behavior in emotional stress group and increase the autonomous activity and the body weight for the rats.
