Objective: To investigate the expression of the miR-23a and Metastasis suppressor1(MTSS1) in colon carcinoma and their clinical significances. Methods: miR-23atargeting MTSS1was evaluated by luciferase reporter vector. Cell growth wasevaluated by MTT. Cell invasion potential was evaluated by transwell invasion assay.And adjacent comparison, hybridization in situ and immunohistochemistry were usedto detect the miR-23a and MTSS1expression in collected colon tissue specimensfrom92cases patient.Results: MTSS1is a target gene of miR-23a. miR-23a downregulated the expressionof MTSS1protein and promoted colon carcinoma cell growth, invasion andmetastasis. The positive expression rate of the miR-23a and MTSS1were86.96%,17.39%in respectivly colon carcinoma,(P<0.01). The up-regulation of miR-23aexpression was associated with advanced clinical stage and the depth of infiltration,and the expression of miR-23a was higher with lymph node metastasis than withoutlymph node metastasis in colon carcinoma. But the down-regulation of MTSS1expression was associated with advanced clinical stage and the depth of infiltration,the expression of MTSS1was lower with lymph node metastasis than without lymphnode metastasis in colon carcinoma. The expression of miR-23a had a close negativecorrelation to that of MTSS1in colon carcinoma (P<0.05).Conclusions:1. miR-23a would promote colon carcinoma cell growth, ivasion and metastasis byinhibiting the expression of targeted gene MTSS.2. miR-23a expression was upregulated in colon cancer and had a close positivecorrelation to advanced clinical stage and with lymph node metastasis. 3. MTSS1expression level was downregulated in colon cancer and had a closenegative correlation to advanced clinical stage and with lymph node metastasis.4. miR-23a had a close negative correlation with MTSS1in colon carcinoma. Theymay be important biological markers for malignant transformation in coloncarcinoma invasion and metastasis.
