AimIn order to investigate the effects of migration and invasion and its molecularmechanism in human gastric cancer SGC7901and SGC7901/VCR cells induced bydiallyl disulfide (DADS).MethodsThe effects of DADS for migration and invasion were examined by WoundHealing assay and Transwell assay and the expression of mRNA and protein ofMMP-9and TIMP-3were determined by RT-PCR and Western blot in SGC7901andSGC7901/VCR cells, respectively.ResultsWound Healing assay discovered that the scuffing distance of SGC7901andSGC7901/VCR cells exposed to15mg· L-1DADS for12h and24h was significantlyincrease than no-treatment group, indicating DADS can inhibit migration in SGC7901and SGC7901/VCR cells (P<0.05).Transwell assay showed that the number of SGC7901and SGC7901/VCR cellsmigrated through the Matrigel treated by15mg· L-1DADS for12h and24h were31.9±0.46and23.5±0.52,obviously decrease than51.6±0.68and41.3±0.72ofno-treatment group; the SGC7901/VCR cell treated by15mg· L-1DADS for12h and24h were36.6±0.58and30.2±0.36,while the no-treatment group were56.9±0.37and45.4±0.71.respectively, indicating DADS can inhibit invasion in SGC7901andSGC7901/VCR cells (P<0.05).RT-PCR showed that expression of MMP-9mRNA was down-regulated andexpression of TIMP-3mRNA was up-regulated in SGC7901and SGC7901/VCR cells treated by15mg·L-1DADS for12h and24h, respectively.Western blot demonstrated that the expression of MMP-9protein was increaseand the expression of TIMP-3was decrease in SGC7901and SGC7901/VCR cellsexposed to15mg· L-1DADS for12,24and48h in a time-dependent manner,respectively.ConclusionDADS may inhibit the migration and invasion in human gastric cancerSGC7901and SGC7901/VCR cells related to the down-regulation of MMP-9andup-regulation of TIMP-3.
