Objective: To create rat model with CP/CPPS, then to observe theexpression of MMP-2、TNF-α in the prostate of these rat model withCP/CPPS. To compare the expression with normal rat and model rat.Method: To choose30healthy, clean, weight about500g adult maleSD rats. To randomly divide them into two groups, the normal controlgroup and the CP/CPPS model group. The normal control group receivedno treatment, feeding normal. The CP/CPPS model group were castratedunder sterile conditions, gentle operation, anatomy carefully, pay attentionto homeostasis, then injected estradiol benzoate (0.25mg/kg/day) for4weeks. After1month, complete remove the prostate tissue of all the rats,then make the prostate by HE staining. To observe these prostate withlight microscope, use the means of immunohistochemistry (sp) to detectthe expression of MMP-2、TNF-α in rat prostatic tissue. Results: Under light microscopy there is no significant ofinflammatory response reaction in the prostates of the control group, whilethe CP/CPPS group have visible significant of inflammatory reactionresponse. After the immunohistochemistry there is negative expression ofMMP-2、TNF-α in normal control group but strongly positive expression inthe CP/CPPS model group. The expression in the two groups is different,the expression of MMP-2、TNF-α in normal rat is lower than that inCP/CPPS model rat(p<0.05).Conclusion: In the pathogenesis and development of CP/CPPS,MMP-2、TNF-α play an important role, and this discovery may be a newtreatment for CP/CPPS. Objective: On the basis of the results of the first part of the study,through injecting testosterone propionate treatment observe the changes inmodel rat with CP/CPPS, to explore a new method in clinical treatment.Method: To choose45healthy, clean, weight about500g adult maleSD rats. To randomly divide them into three groups, the normal control group, the CP/CPPS model control group and the model treatment group(CP/CPPS model+testosterone propionate treatment group). The modelcontrol group and model treatment group were created CP/CPPS model.Then model treatment group were injected testosterone propionate1mg/kg/d, while the other two groups were injected the same amount of NS.7days later kill all the rat, complete remove all the prostate tissue, thenmake the prostate by HE staining. To observe these prostate with lightmicroscope, use the means of immunohistochemistry (sp) to detect theexpression of MMP-2、TNF-α in rat prostatic tissue.Results: To observe the tissue under the microscope. There is noinflammatory infiltration in the control group, the model control grouphave obvious inflammatory infiltration, while the model treatment grouphave little inflammatory infiltration. The expression of MMP-2、TNF-α inthe model groups was significantly higher than that in the normal controlgroup(P<0.05);(3)The expressions ofMMP-2、TNF-α inthe testosteronetreated group were significantly decreased(P<0.05).Conclusion: The expression of MMP-2、TNF-α in model controlgroup is significantly higher than the other two group, while give thetreatment with testosterone propionate the expression decreasedsignificantly. This result suggest that testosterone propionate could treat theCP/CPPS, testosterone could be a new treatment option of CP/CPPS byinhibiting the expression of MMP-2and inflammatory cytokines(TNF-α)....
