The aim of the study was to promote the standardization of Codonopsis pilosula (Franch.) Nannf, and to provide a basis for germplasm identification,seed testing and seed production so that the problems such as seed disputes and seed quality irregularity due to the lack of seed standard could be solved. We collected and investigated44seeds from8counties in Shanxi, Gansu and Sichuan province. Referring to<The Rules for Agricultural Seed Testing>(GB/T3543-1995) issued by China, the seed quality of codonopsispilosula from different areas was measured. The seed testing methods for quality items, including sampling, purity analysis, germination test, verification of genuineness, determination of moisture content, seed health test, seed viability test andweight determination of codonopsispilosula had been initially established. According to the rules, we tested the quality inspection, statistical analysis of new harvest of No.25-43seed samples and put forward the standards for Codonopsis pilosula (Franch.) Nannf seed.Based on the research of testing ways we concluded:Sampling was taken with the method of coning and quartering. The total weight of submitted sample was100g,among this, lOg for purity analysis, lOg for germinative testing,20g for authenticity identification, and50g for moisture content. The least weight of working sample for purity analysis was1.5g. The pure seeds, impurities and other seeds were isolated repeatedly by testing sieve.; the best germination condition was20℃,2400lx and paper bed; The most suitable method for determination of moisture content was high temperature ground drying method with drying time of5h.The seed viability test was taken by TTC, With the condition of seed perforation,1%concentration of TTC,40℃of dyeing temperature and3h of dyeing time. The seed should be perforated before immersing in TTC. One thousand grains’s way was taken to test the weight; seed health test was taken with PDAMedium,we choose seeds’ protain SDS—PAGE electrophoresis to verify the genuineness.According to the test of Codonopsis pilosula (Franch.) Nannf we concluded:The purity of the seed was very weak. Its main inert matter of were gravels,so we should clean seeds before saving them in case of seeds wearing. Storage time and conditions also influenced seed germination rate greatly, so we should store the seeds in20℃low-temperature environment and the longest storage time was1year. The seeds’moisture content was so low that it was apt to be dried; The predominant bacterium that Codonopsis pilosula (Franch.) Nannf carried was Fusariumlink which is the main pathogen to cause root rot, so we should disinfect the seeds before sowing them. According to the analysis of the result we concluded the genetic relationship among seeds from Fengxian in Shanxi, Wenxian in Gansu and Aba in Sichuan provinces were more closed. The molecular weight of distinct protein was170KDa. From the result of grading we could find the quality of seeds from Shanxi was better than Gansu. |
