| Objective: To study the association between angiotensinogen (AGT) geneM235T polymorphism, angiotensin Ⅱ type1receptor (AGTR1) gene A1166Cpolymorphism, aldosterone synthase(CYP11B2) gene-344T/C polymorphism and therisk of Ischemic Storke in China southern han crowds.Methods: AGT gene M235T polymorphism, AGTR1gene A1166C polymer-phism, CYP11B2gene-344T/C polymorphism were genetyped in129cases of IS and81the controls by PCR and gene sequencing, and contribution of the three genes tothe risk of IS was analysed by binary logistic regression analysis. The IS complicatedwith hypertension group is compared with the normotensive controls to study thecorrelation between the three genes and the IS complicated with hypertension group.Combined with clinical data, difference of clinical data of various genetype of AGT,AGTR1and CYP11B2is analysed by ANOVA.Results: The frequencies of the AGT gene TT genotype and T allele in the ISgroup is not significantly different with those in the control group(χ~2=2.196, P=0.334and χ~2=1.719, P=0.190). The frequencies of the AGTR1gene CC genotype and Callele in the IS group were not significantly different with those of the controls(χ~2=1.612, P=0.566and χ~2=1.587, P=0.208). The frequencies of the CYP11B2gene CCgenotype and C allele in the IS group were not significantly different with those ofthe controls(χ~2=2.752, P=0.253and χ~2=0.023, P=0.880). Adjusted for age, gender,BMI, hypertension, diabetes mellitus, smoking history, dyslipidemia, Binary logisticregression analysis shows that association between AGT or AGTR1and the risk ofthe IS was observed but no statistical significance.(P=0.113and0.082). Adjusted forage, gender, BMI, diabetes mellitus, smoking history, dyslipidemia, Binary logisticregression analysis shows that association between AGT and the IS complicated withhypertension group is statistically significant(Waldχ~2=4.624, P=0.032, OR=2.340).Binary logistic regression analysis shows that the OR of MM genotype is8.148 (95%CI:0.816-81.353)(P=0.074). For all210cases, The study shows that total chole-sterol levels of the crowds with TT genotype of AGT gene is significantly greaterthan the crowds with the MT genotype(P=0.038). Comparison of the Glucosebetween the crowds with the AA genotype of the AGTR1gene and the AC genotypegroups tend to be statistically significant (P=0.051). Comparison of the totalcholesterol levels between the crowds with the CT genotype of the CYP11B2geneand the crowds with the TT genotype tend to be statistically significant(P=0.077).Conclusion: The AGT gene M235T polymorphism is probably associated withthe risk of Ischemic Stroke complicated with hypertension in the southern han crowds.and AGTR1gene A1166C polymorphism are probably associated with IschemicStroke and Metabolic syndrome. CYP11B2gene-344C/T polymorphism is notrelated to Ischemic Stroke in the southern Han crowds. These results need to beconfirmed in a larger cohort. |
