Objective We combined three absorbable materials with PP meshes to make newcomposite patches and then observe the local inflammation, tissue integration andanti-adhesions of this three new domestic composite patches repairing abdominal walldefect in dogs under contaminated environment, and to investigate the characteristics of3composite patches and the feasibility of one-stage repair.Methods Twenty-four adult male Dogs (weighing16-20kg) were used in thisresearch three contaminated defect models were set up on the up, left and rightabdominal wall of one dog, and three different patches were randomly divided into threedifferent defects to complete the repair. We divided three different patches into threegroups: PP/PLC composite patches (group A),PP/HA/PLC composite patches (groupB), and PP/collagen/PLC composite patches (group C). After the wall defect model hadbeen established, we flled small intestine with10ml saline0.9%solution, thenaspirated and spilled on the abdominal cavity, the abdominal wall and the mesh, at lastwe used3different patches, according groups (size5×5cm), to enclose the defect. Theordinary condition of all animals had been recorded after surgery. At90dayspostoperatively, laboratory animals were sacrificed, the adhesions was observed, and thepatch and adjacent tissue was harvested for histological observation.Results There were no animals died in this experiment at90days postoperatively.Hernias and peritoneal abscesses were not be found at all the animals. According to themodified Katada criteria, no significant difference in the adhesion score was foundbetween Group A and Group B (P>0.05), Group C had a lower score than Group A andGroup B, The histological results showed that on inflammatory cell infiltration, GroupC had a lower score than Group A and Group B, the difference have statistical sense (P<0.05), on fibroblasts and neovascularization, there are no significant differenceamong the three groups (P>0.05). ConclusionAll of the three new domestic patches can use in the contaminated abdominal walldefects, no severe complications and animals death were found after the surgery, onanti-adhesions the composite patch of PP/Collagen/PLC was better than PP/PLC groupand PP/HA/PLC group. On histological observation, inflammation of PP/Collagen/PLCgroup was more light than the other groups, there no difference of the three compositepatches on fibroblasts and neovascularization. Generally speaking, PP/collagen/PLCgroup had got the best results on repair of contaminated defect of the abdominal wall indogs, this new composite patch had a certain clinical application prospect.
