An.minimus is an important malaria vector. Indoor residual spraying and pyrethroidinsecticide treated mosquito nets have been important chemical control methods inYunnan Province. It has been reported that pyrethroid insecticide resistance has beenfound in An.minimus. The blood-feeding behavior and resting behavior are alsodifferent between An.minimus A and An.minimus C. The effects of Insecticideselection pressure on An.minimus insecticide resistance and behavior should befurther study.1. An.minimus molecular typingIt is difficult to distinguish between An.minimus complex and several relatives. Themorphological classification of the collected mosquitoes was finished first, and thenmolecular typing was detected by PCR method.1047mosquitoes had been collected,included364individuals of An.minimus A and593individuals of An.minimus C and19individuals of An.jeyporiensis and14individuals of An.aconitus.2. Study of habitat habits of An.minimusThe sampling points were several cowsheds and human houses in the DongFanghongvillage.The time to capture adult mosquitoes by trapping method was from19:00to8:00the next day.With statistical analysis by spss software, An.minimus A preferhuman house, An.minimus C prefer cowshed.3. Study of Blood-feeding habits of An.minimusMosquitoes can suck blood from cattle, pigs, chickens and people in local area. The primers of PCR were designed to identify collected mosquito stomach blood source.Analysis the result with Spss, there was no obvious blood-feeding preferences in Anminimus A, and An minimus C prefere cattle blood.4. Study of sodium channel gene sequences of An.minimusAn.minimus para-type sodium channel gene can be found in Genbank(GenBank:EU155386.1). The ⅡS6gene was amplified and sequenced. No mutation was foundat the1014th loci based on,which the common kdr mutation loci is. It showed that theknockdown resistance level of An.minimus in this region is very low.5. Results and analysis of the diversity of microsatelliteThere are28microsatellite sequences published in Genbank. The serial numbers areHQ839797.1, HQ839799.1, HQ839801.1, HQ839803.1, HQ839797.1,HQ839795.1, HQ839793.1, HQ839793.1, HQ839789.1, HQ839816.1,HQ839814.1, HQ839810.1, HQ839812.1, HQ839808.1, HQ839806.1,HQ839804.1, HQ839802.1, HQ839802.1, HQ839798.1, HQ839796.1,HQ839794.1, HQ839792.1, HQ839792.1, HQ839815.1, HQ839813.1. DesignedPrimers in accordance with the above sequence. Locus AM4, AM7-1, AM8, AM12,AM21, AM23, AM25-1and AM27-1amplified a clear bands, and the sequencingresults were satisfactory. The seven locis can be used for microsatellite analysis.Mosquitoes were divided into eight groups: Group A was An.minimus A (YunnanDongfanghong Village), Group B was An.minimus C (Yunnan Dongfanghong Village),Group C was An.jeyporiensis (Yunnan Longtang Village), Group D was An.aconitus(Hainan Yaying Village), Group E was An.aconitus (Yunnan Longtang Village),Group F was An.minimus A (Yunnan Longtang Village), Group G was An.minimus C(Yunnan Longtang Village), Group H was An.minimus A (Hainan Yaying Village).Microsatellite markers were used to analyze Ho, He,PIC, Fst, Fis, Fit,Hardy-Weinberg equilibrium analysis. Ho and He were very similar at each locus,suggested that these markers have good polymorphism for analysis. The PIC value isvery good in eight groups of populations. Hardy-Weinberg equilibrium test suggested that loss of heterozygosity appeared at multiple loci, deviated from theHardy-Weinberg equilibrium standard. Fst=0.1386, there is less gene flow betweenthe various groups. The cluster analysis showed that the proximity between the samespecies. An.minimus A and An.minimus C were proximity with An.aconitus,An.jeyporiensis was farther. This was in line with the basic situation of theAn.minimus group.
