Objective:To establish a new accurate, sensitive and simple electrochemical method for the determination of vitamin K3in serum,and to determination the Medical reference range of VK3in serum by LK3000V electrochemical workstation. To synthesize the nano silver,and determinate the trace amounts of Ag+by a glassy carbon electrode coated with film of4-tert-Butyl-1(Ethoxycarb only methoxy) Thiacalix[4]Arene(TAC).Methods:1、VK3:There was a three electrodes system with glassy carbon hydrarg dressed as working electrode, Ag/AgCl as reference elect-rode, Pt as auxiliary electrode. With the three electrodes system, the content of vitamin K3standard solution was determined by the method of differentinal pluse stripping voltammertry in NH3·H2O/NH4CI buffer which pH was9.4and concentration was1.0mol·L-1;2、Ag+:Silver nano-particles was synthesized by the way of second reduction and there was a three electrodes system with glassy carbon TAC dressed as working electrode, saturated calomel electrode as reference electrode, Pt as auxiliary electrode. With the three electrodes system, the content of vitamin Ag+standard solution was determined by the method of differrentinal pluse stripping voltammertry in B-R buffer which pH was4.5.Results:1、 A good linear relationship between the stripping peak current and vitamin K3concentration in range of0.23-1.6ng·mL-’was held (r=0.9994), with a detection limit of0.23ng·mL-1. RSD was1.64%for the determination of vitamin K3at level of0.92ng-mL-1(6times). The recovery rate was99.91%, I n the process of impleme-ntation of the program, Tianjin Nankai Hospitaland the Tianjin Municipal People’s Hospital, two centers were completed158cases and162cases, the number of cases of age group distribution balance, no significant difference between the design requirements to achieve the clinical trial program; LK2006electrochemical the test results of the workstation95%medical reference range of0.1to1.2ng·mL2、The nanoparticle is small and evenly distributed and with no apparent gather. A good linear relationship between the stripping peak current and vitamin K3concen-tration in range of2.5-40ng-mL-1was held (R2=0.994), with a detection limit of2.5ng-mLL-1. RSD was1.01%for the determination of vitamin K3at level of20ng-mL-1(6times).Conclusion:1、Compared with the method of high pressure liquid chromat-ography (HPLC), it is simple rapid、sensitive, and it can be a high sensitive method for the determination of vitamin K3in serum.2、Because of its high specificity, sensitivity and other characteristics, less can be used as the Ag+sensitive detection methods; the system the stability of nano silver particles, dissolution of Ag+can increase the bactericidal effect.
