| Objective:To study the expression of coxsackie and adenovirus receptor (CAR) mRNA and protein in in EGFR mutant and EGFR wild-type of human NSCLC,analyzing their correlation with EGFR gene mutation and CAR interaction mechanism in the tumor development process. To analysis the EGFR-TKIs treatment of EGFR mutant NSCLC possible mechanism and to provide a theoretical basis for the common EGFR-TKIs and adenovirus vector therapy of tumors.Method:Collecting fresh non-small cell lung cancer300samples,put into liquid nitrogen as soon as removed from the patient’s body,80℃followed. Simultaneously the clinical data of the patients was collected for the results analysis. First,using real-time PCR detect the gene mutant state of19and21of the above sample. Randomly selected50cases for the mutation group and50cases of wild group to determined the expression of CAR and pEGFR protein was by IHC and Western blot.Analyzing CAR expression in EGFR mutation group and wild group and investigating the interrelation and effect of the two.Results:1. EGFR gene mutation state102samples were mutation group and198were wild type group.The mutation focus on exon19and21The samples of Exon19were66and21wre34.2.The expression of CAR and pEGFR protein:Randomly selected50cases for the mutation group and50cases of wild group, IHC result shows70(70%) cases were CAR positive, of which48(96%) cases were mutation samples and22(44%) were wild type samples.69(69%) cases were CAR positive, of which49(98%) cases were mutation samples and20(40%) were wild type samples.By Western blot assay, a band of about46kd was detected in mutation group and wild group.with the expression value0.891±0.144,1.220±0.269. The expression value of pEGFR protein were0.891±0.144,1.220±0.269in mutation group and wild group.The expression value of CAR and pEGFR has significance between mutation group and wild group (P<0.05). 3. The expression of CAR protein and EGFRprotein has correlation with EGFR gene mutation.but there is no correlation between the two with the type of EGFR gene mutation,P>0.05.4. The correlation of CAR expression with pEGFR proteinin EGFR mutation group and wild group.There was a statistically positive correlation between expression of CAR protein and pEGFR(r=0.448, P=0.026).Conclusion:1. Gene mutation and pEGFR expression:EGFR mutation behaves on exon19and21, Exon19is more than21.2. The expression level of CAR protein and EGFR protein has significant difference among in mutation group and wild group of NSCLC.3. CAR expression has significantly positive correlation with p-EGFR and EFGR gene mutation state in NSCLC. But there is no correlation between the two with the type of EGFR gene mutation.4. If CAR expression and EGFRgene mutation play concomitance role on tumor formation, we might apply the antibody-directed therapy as EGFR TKI assistance in human lung cancer. If CAR is not a promoting factor of human lung cancer, the elevated expression of CAR in lung cancer will be more favorable to the gene therapy of human lung cancer. and advise theory base for combining therapy of EGFR-TKIs and ada-mediated gene therapy. |
