objectives:To study on effects of Fengdan in type2diabetic mice, identifies the main bioactive components of fengdan,and preliminary study on its mechanism of its action of lowering blood, providing theoretical basis for clinical application of Fengdan.Method:Continuous feeding high fat diet (lard10%, sugar20%, cholesterol2.5%, bile salt1%, common feed66.5%)4weeks to establish male Kunming mice Hyperlipidemia model, Then through intraperitoneal injection of Streptozotocin (STZ)130mg·kg-1to establish the experimental mouse model of type2diabetes. First, boiling Fengdan with10times the amount of water three times,2hours for each time. To merger three times filtrate after filtered with gauze and filter paper every time then evaporating filtrate. Adding with alcohol volume of95%ethanol in concentrated liquid to the final containing alcohol volume of30%,store in4℃of refrigerator for24hours, then filtered. The small molecular group was the filtrate and the polysaccharide group was the residue. The Diabetes mice was randomly assinged to the metformin group, the group treated with Fengdan polysaccharides and the group treated with FengDan small molecular. Continuously intragastric admini-stration for4weeks,body weight were recorded weekly. The levels of blood glucose concentration (GLU), glucose tolerance,total cholesterol (TC), three benzoyl peroxide glycerol (TG), and other biochemical indexes were determined by semi-automatic biochemical analyzer,and the levels of hydroxymethyl glutaryl coenzyme A reductase (HMG-CoA), glucose-6-Phosphatase (G6PC), and glucokinase (GCK) were determined by real time PCR.Diabetes mellitus (DM, Diabetes Mellitus), one of the most common chronic and metabolic disease in the world, characterized by chronic high blood glucose along with disorders of glucose, fat and protein metabolism caused by defect of secretion of the insulin. The high blood glucose as the main indexes in Clinic, more systems will be damaged caused by long illness. The prevalence of diabetes increased as the population age increases, improved medical conditions make the diagnosis of diabetes mellitus in the elderly more quickly, along with the improvement of living standards and population overall life extension, the total number of diabetes will also further increased. Looking for more high effective, stable and durable, lower side effects of drugs and treatment methods, become the focus of researching diabetes in recent years.RESULTS:1fengdan make a great improvement in the spirit of the diabetic mice, increase its intensity of activity, weight gain, and improve the clinical symptoms.2Fengdan can significantly reduce blood glucose of the diabetic mice, improve glucose tolerance, decrease serum total cholesterol and triglyceride levels. The fengdan polysaccharide composition is the most effective. 3Fengdan can significantly enhance the content of superoxide dismutase and can significantly reduce the content of malondialdehyde, enhances the antioxidant ability of the diabetic mice.4Fengdan can significantly reduce glucose-6-Phosphatase (G6PC) and hydroxy-methyl-glutaryl Coenzyme A reductase (HMG-CoA reductase) and can significantly increase glucokinase (GCK) expression, promoted the glucose metabolism in diabetic mice.5The Fengdan polysaccharide components is the best effective hypoglycemic of Fengdan in the diabetic mice. The hypoglycemic mechanism may be promotion the biosynthesis of pancreatic islet Pcells and promotion glucose metabolismConclusions:In the present study found that Fengdan can significantly improve the glucose and fat metabolism of the2type diabetic mice, reduce glucose toxicity and fat toxicity, and prevention the development of diabetes and its complications. Clearing the hypoglycemic component is the best bioactive in hypoglycemic. Open up new ideas for the traditional Chinese medicine in the treatment of diabetes. In addition, fuether comprehensive pharmacologic investigations, including experimental long-term studies, should be carried out on Paeonia suffruticosa Andr.or its hypoglycemic mechanism.
