Objective: To investigate the correlation between the polymorphisms (rs931127andrs746429) in signal-induced proliferation-associated1(Sipa1) gene and circulatingtumor cells in breast cancer patients.Methods: The polymorphisms of Sipa1were detected by PCR-DNA sequencing in124primary breast cancer patients and120healthy controls. The circulating tumorcells were detected by real-time quantitative PCR.Result: The polymorphisms weren’t associated with the circulating tumor cells of thebreast cancer patients (P>0.05). rs931127and rs746429polymorphisms of Sipa1were not significantly associated with patient age, estrogen receptor status,progesterone receptor status, levels of C-erbB2over expression (P>0.05). However,patients with the rs931127homozygous for the wild-type GG genotype were lesslikely to have positive lymph node involvement (22.6%) than patients withheterozygous GA (48.3%) and mutant AA genotype (71.0%)(P=0.002). Patientswith the rs746429homozygous for the wild-type CC genotype were less likely to havepositive lymph node involvement (31.4%) than patients with heterozygous CT (64.2%)and mutant TT genotype too (44.4%)(P=0.003).Conclusion: The variant rs746429and the rs931127allele did not associate with thecirculating tumor cells of the breast cancer patients. However, they were associatedwith lymph node involvement. Breast cancer patients with rs931127A allele andrs746429T allele may be associated with lymphatic metastasis.
