Objective: through detecting the expression levels of signal transducerand activator of transcription factor STAT5mRNA and STAT5protein innon small cell lung cancer (NSCLC) in blood of the patients, toinvestigate its effect on diagnosis for non small cell lung cancer, and toprovide new index for the clinical diagnosis of early stage lung cancer.Methods:â‘ using ordinary PCR method to detect the expression ofSTAT5mRNA in peripheral blood of15patients with NSCLC,15cases ofpulmonary benign space-occupying lesions of patients and15cases ofnormal human, and compared with its differential in different groups.â‘¡enzyme-linked immunosorbent assay (ELISA) detected60cases ofnon-small cell lung cancer patients,60cases of pulmonary benignspace-occupying lesions,60cases of healthy people STAT5protein andtumor markers in serum CYFRA21-1, of CA125, CEA expressionanalysis serum of STAT5protein levels in NSCLC clinical andpathological factors, and ROC curve to evaluate and compare STAT5andtumor markers of CYFRA21-1, the diagnostic value of CA125, CEAsingle seizure, and the joint of CYFRA21of STAT5protein-1, of CA125, CEA diagnostic value of NSCLC. Results:â‘ PCR to detect the ofSTAT5expression of mRNA in these samples of the3groups the resultsby the homogeneity of variance test between the two groups werecompared using single factor analysis of variance Tip: significantlyhigher expression of STAT5mRNA in lung cancer group pulmonarybenign space-occupying lesion group and the healthy control group (P<0.01); lung benign space-occupying lesions and the healthy controlgroup expression differences not statistically significant (P>0.05)2ELISA, test results showed STAT5protein in the lung cancer group,benign lung space occupying lesion group and the healthy control groupwere expressed. Tip: The one-way ANOVA statistical analysis of STAT5protein content in the lung cancer group were significantly higher than inbenign pulmonary placeholder group and the healthy control group (P<0.01); benign space-occupying lung disease patients and healthy controlgroup of STAT5protein content was no statistical difference significance(P>0.05).â‘¢lung cancer group, the serum STAT5protein content in theadenocarcinoma group was significantly higher than the carcinoma group(P <0.05);, no significant differences in STAT5protein content in thepoorly differentiated group (P>0.05); of STAT5protein in the differentgender groups, the difference is not statistically significant (P>0.05);with lymph node metastasis and without metastasis content difference notstatistically significant (P>0.05)(3) serum STAT5protein and CYFRA21-1, CA125CEA single prosecution under the ROC curve area(Az) for0.811,0.775,0.789,0.792four Az are in the range of0.7to0.9,the diagnostic value are medium, using the Z test showed no significantdifference (P>0.05)â‘£of CYFRA21-1, CA125, CEA and three joint testROC curve area0.856; increased the STAT5four joint survey under theROC curve area of0.923. Az area of two programs by the Z-test prompt,the difference was statistically significant (P <0.01) Conclusion:(1)STAT5mRNA in non-small cell lung cancer group, lung benignspace-occupying group group and the healthy control group, serum wereexpressed in non-small cell lung cancer patients showed abnormally highexpression, the expression level is higher than in benign pulmonarydisease group and the normal comparison group.â‘¡The serum of STAT5protein expression in non-small cell lung cancer patients, benign lungdisease patients and normal blood, and in non-small cell lung cancerpatients showed abnormally high expression, the expression levels andtumor pathological type, in which STAT5The protein expression ofgender-related, regardless of the degree of tumor differentiation andstaging.(3) of STAT5protein can reach the current recognized andcommonly used clinical tumor markers CEA, CA125and CYFRA21-1non-small cell lung cancer, the same diagnostic value, indicating thatSTAT5as NSCLC markers to help diagnose potential.â‘£After the tripleand quadruple prompt increase in STAT5, you can increase the sensitivity and accuracy of detection of NSCLC. Further illustrate the diagnosticvalue of STAT5in NSCLC.
