| Objective:The relationship of different pregnant rats super-dose of food additives on fetal rat development, the impact of the placenta and fetal rat brain tissue c-fos gene expression.Explore the super-dose of the combination of food additives on the different stages of gestation fetal rat embryo toxicity, the manifestations and the relationship between fetal rat brain c-fos gene expression.Method:The establishment of pregnancy combined with ultra-dose food additive model, using HE staining and reverse transcriptase polymerase chain reaction to detect pregnancy fetal rat placental tissue, the pathological changes of brain tissue and the expression of c-fos gene in the hippocampus.Result:(1) The ratio between absorption fetus and dead fetus group A (applying overdose food additive for2-7days) is15.58%(12/77). The ratio between absorption fetus and dead fetus in group B (applying overdose food additive for8-13days) is4.87%(4/82). Comparing with group C (applying overdose food additive for14-19days), which is1.22%(1/83). The still birth rate is obviously rising. There are significant differences (P<0.01, P<0.05). Comparing the ratio between absorption fetus and dead fetus of group A and group B with group D (control group), which is1.14%, there are similar results (P<0.01and P<0.05). but there is no significant difference between group C and group D.(2) The average weight of embryo mice and average placenta weight in group A is3.75±0.12mg,0.593±0.183mg, it is3.42±0.10mg,0.574±0.348mg respectively in group B. it is notably lower than the average weight of embryo mice and placenta in group C, which is4.01±0.09mg,0.623±0.214mg. There are differences (P<0.05). Comparing the average weight of embryo mice and placenta in group D, which is4.12±0.43mg,0.654±0.212mg. there are similar results (P<0.05). There is statistical meaning comparing with group A and group B (P<0.05). There is no difference between group C and group D (P>0.05).(3) The average body length and tail length is3.47±0.14mm,1.02±0.56mm. There are statistical meanings comparing with other groups (P<0.05). There are no differences in other groups.(4) the IUGR rate in group B is28.2%(22/78), it is strikingly higher than other three groups (group A is8/65=12.3%, group C is3/84=3.57%, group D is1/86=1.16%). There are differences (P<0.05). Comparing the IUGR rate in group A with group C and group D, There are differences (P<0.05). There is no difference of the IUGR rate between group C and group D (P>0.05).(5) The pathological change of the placenta and the brain tissue is remarkable in group B.(6)Comparing the brain tissue c-fos gene expression in group B with other three groups, there are statistical meaning (P<0.05).Conclusion:Applying overdose food additives to mice during pregnancy, can lead to embryotoxicity, the effect extent is related with duration of pregnancy. Pregnancy super-dose of food additives can cause pregnant rat placental tissue and brain tissue pathological changes during8-13days. Pregnancy super-dose of food additives can cause fetal rat brain c-fos mRNA overexpression, it is also remarkable during8-13days. This is another rat pregnancy8-13days during early pregnancy is an important time window of super-dose of food additives on the brain tissue damage.As for how the super-dose combination of food additives cause changes in brain tissue and the mechanism is subject in the future. |
