The root of Dictamnus dasycarpus which belongs to Rutaceae with the effectiveness of heat and detoxification. The root of D. dasycarpus can treat eczema,skin itching and hepatitis. The root of D. dasycarpus has been used widely for a long history in various regions of China. Its unique scientific value deserves to be explored.The topics in our paper mainly focused on chemical composition. We investigated that the compouds isolated from D. dasycarpus would have effects on Hep G2cell line. Finally, we reviewed the protective of D. dasycarpus on D-galactosamine/LPS induced acute liver injury on mice.In this research, D. dasycarpus was extracted by95%ethanol, and then was suspended in water and successively partitioned and extracted with petroleum ether, ethyl acetate and n-butanol. The active sites of the three was separated by different chromatographic methods, fifteen compounds were isolated from D. dasycarpus, their structures were elucidated by their physical and chemical properties and NMR techniques. Their structures were identified as β-sitosterol (1), Dictamine (2), Preskimmianine (3), Glycolone (4), Limonin (5), Fraxinellone (6), Obacunone (7), Evdol (8), Fraxinellonone (9) Scopoletin (10)7-isopentenyloxycoumarin (11),11,12,13-trinorguai-6ene-4β,10β-diol (12),5,6,7-trimethoxycomarin (13), Skimmianine (14), Daucosterol (15). Glycolone (4),7-isopentenyloxycoumarin (11) and5,6,7-trimethoxycomarin (13) were isolated from the plant for the first time.In this part of the paper, we establish six teams of the experiment. They are the negative team with no drugs, the t-BHP-induced team, and compound-treated team with concentration10μM,20μM,40μM,80μM, resp. We screened out2compounds which have hepatoprotective effects on t-BHP-induced cytotoxicity in HepG2cells from D. dasycarpus. by MTT method. The result of experiment is that the cell viability of Obacunone EC50was53.23H.M. Based on the appeal results, Obacunone could reduce the cytotoxicity observably. |
