Objective:To analyze the difference of microtubule associated protein Tau expression between nasopharyngeal carcinoma parental cells and paclitaxel-resistant cell lines, investigate the role of Tau in paclitaxel-resistance in nasopharyngeal carcinoma.Method:Use nasopharyngeal parental cell lines of CNE-1, CNE-2and its paclitaxel-resistant cell lines CNE-1/taxol, CNE-2/taxol as experimental subjects, detect the expression of Tau protein and mRNA by Western Bolt, and real-time fluorescence quantitative PCR respectively.Results:The results of Western Bolt, and Real-time quantitative PCR reflect the Tau protein and mRNA expression in different cell lines. The relative optical density of reference value of the Tau protein/GAPDH reflect the expression level of Tau protein in different cell lines. mRNA expression is calculated by the method of2-ΔCt. The results showed that all the NPC cells in experiment expresses Tau protein, and Tau protein in paclitaxel-resistant cells(CNE-1/taxol, CNE-2/taxol)is higher than parental cells(CNE-1,CNE-2), respectively3.11±0.54,1.39±0.21times. Tau mRNA also expressed in both NPC parental cells and paclitaxel-resistant cells, and in paolitaxel-resistant cells (CNE-1/taxol, CNE-2/taxol) is2.48±0.33,1.67±0.15times higher than NPC parental cells(CNE-1,CNE-2).Conclusion:The expression of Tau in NPC paclitaxel-resistant cell lines is higher than the parental cell lines. Tau may be associated with paclitaxel resistance. Tau can be used as a molecular to predicted paclitaxel Chemotherapy sensitivity and prognosis. Intervention of Tau could reverse paclitaxel-resistance to some extent. |
