| Background and aim of the study:The purpose of this experi-ment is to investigate whether HCPT inhibit the Raji cells and induce them apoptosis and the relation between Caspase-3proteins and this apoptosis.Methods:(1)MTT assay was used to investigate whether HCPT inhibit the Raji cells.(2)Flow cytometric analysis were used to investigate whether HCPT induce Raji cells apoptosis.(3) Immunohisto-chemistry were used to investigate the relation between apoptosis and Caspase-3proteins.Results:(1)HCPT can inhibit the Raji cell,when the drug at the concentration of0-10ug/ml,the HCPT and its inhibit rates got three line correlation coefficient(r) in24ã€48ã€72h.The r were:0.946ã€0.887and0.881; The P were:0.015ã€0.045and0.048.A11the P<0.05.The inhibit rates(IR) increased with HCPT concentration;When the concentration of HCPT is5ug/ml,the IR increased with treat time,and the IR increased with treat time when the time range is from24-48h as other HCPT concentration.(2)HCPT can induce Raji cell apoptosis,the apoptosis rates were significant different with non HCPT treat group and HCPT treat groups in24h and72h(P<0.05).(3)The Caspase-3proteins in the HCPT treat group is higher than the nonHCPT treat group (t=-5.04,P<0.01). Conclusions:(1).HCPT can inhibit the Raji cells,In the range of concentration is from0-10ug/ml,the inhibit rates(IR) increased with HCPT concentration; When the concentration of HCPT is5ug/ml,the IR increased with treat time,and the IR increased with treat time when the time range is from24-48h as other HCPT concentration.(2).HCPT can induce Raji cells apoptosis,the rate of apoptosis is the largest when the concentration and treat time were propoer.(3).The Caspase-3proteins in the HCPT treat group is higher than the non-treat group,the apoptosis maybe induced by the Casebase-3proteins increased after treat by the HCPT. |
