OBJECTIVE:The present study is to establish the separation of lignans method in the dried flower buds of Magnolia sprengeri Pamp. and a measurement to test the content of5kinds of lignans in M. sprengeri, in order to improve the quality evaluation of M. sprengeri.METHOD:On the Based objective of the present study, the relevant literatures of Magnoliae Flos were first collected. Isolation and purification of lignans from M. sprengeri by silica gel column chromatography and high-speed counter-current chromatography.The structures of the compounds were identified by ESI-MS and NMR.Then a multi-assessment method to measure the quality evaluation of M. sprengeri was set up. In order to establish a new method of quality evaluation and validate its feasibilities by the simultaneous quantitative assay in M. sprengeri, the new qulity evaluation method, quantitative analysis of multi-components by single marker (QAMS), was established and validated with M. sprengeri. We used (-)-maglifloenone, futoenone, fargesone C, fargesone A, licarin B as analytes to evaluate the quality. Then10batches of samples, collected from different drug markets, were determined by both external standard method and QAMS and found no significant differences between external standard method and QAMS. After compared the results of external standard method evaluation with a multi-method test results, QAMS is feasible and suitable to evaluate the quality of M sprengeri. RESULTS:With the method of silica gel column chromatography and high-speed countercurrent chromatography,11lignans were successfully isolated from M. sprengeri. All of these compounds were elucidated as (-)-maglifloenone, futoenone, magnoline, cylohexadienone, fargesone C, fargesone A, fargesone B, licarin B, acuminatin, kadsurin A and fargesone D. We measured using a multi-assessment method and external standard method for the determination of the10batches of M. sprengeri (-)-maglifloenone, futoenone, fargesone C, fargesone A, licarin B, the test showed that no significant difference between the two methods.CONCLUSION:The research work and the results obtained, and further improve the quality evaluation of M.sprengeri.
