Research Of Different Expession Of HSP60in Docetaxel Treatment Of Advanced Hormone-Resistant Prostate Cancer

ObjectiveThe current study utilized the treatment of Docetaxel to the advanced hormone-resistant prostate cancer. Apoptosis and docetaxel-resistant PC-3cells were induced by different concentrations of Docetaxel.And the proteomics were used to identify the differentially expression of heat shock protein60(heat shock protein60, HSP60) in apoptosis and docetaxel-resistant PC-3Cells from the docetaxel-sensitive PC-3cells, and investigated the possible role of HSP60in the process of apoptosis and resistance of PC-3cells formed.MethodFirstly, PC-3cells were respectively disposed by10uM/L、20uM/L、30uM/L40uM/L and50uM/L Docetaxel, and the SRB and Hoechst staining were used to synchronously detect the proliferative inhibition and the apoptosis induction of Docetaxel to the PC-3cells. According to the results, Apoptotic PC-3cells were induced and produced by20μM Docetaxel.Secondly Docetaxel-resistant PC-3cells were produced by breeding with increasing dose of docetaxel gradually. Then2D-DIGE was used to get the protein expression profile between untreated PC-3cells and Apoptotic PC-3cells, docetaxel-sensitive PC-3cells and Docetaxel-resistant PC-3cells,and differentially expressed proteins were quantatively screened by the comparison of protein expression profile. matrix-assisted laser desorption/ionizafion time of flight mass spectrometry(MALDI-TOF-Ms) was used to get the peptide mass fingerprints(PMFs) of the differentially expressed proteins.PMFs were searched by the Mascot software.And the differential expressions of HSP60between docetaxel-sensitive PC-3cells and Apoptotic PC-3cells, docetaxel-sensitive PC-3cells and Docetaxel-resistant PC-3cells were finally detected.ResultsBy the searching result of Mascot software for the PMFs, the expression of HSP60was up-regulated in Docetaxel-induced apoptotic PC-3cells, and down-regulated in Docetaxel-resistant PC-3cells.The differences are both greater than2times.Conclusion(1) The expression of HSP60was up-regulated in Docetaxel-induced apoptotic PC-3cells, and it suggest that the over expression of HSP60may lead to the apoptosis of PC-3,or the apoptosis of PC-3lead to the over expression of HSP60.(2) The expression of HSP60was lowly expressed in Docetaxel-resistant PC-3cells.and it suggest that the low expression of HSP60may play a role in the process of the resistance formed,or the resistance of PC-3lead to the low expression of HSP60. According to these results,HSP60may be as prognostic indexes for Docetaxe treatment to the advanced hormone-resistant prostate cancer, and it provide an experimental evidence for researching the effect of HSP60in the development of advanced androgen independent prostate cancer. cancer.
